    Gamma Secretase Inhibitors, DAPT and MK0752, Exhibit Synergistic Anticancer Effects with Cisplatin and Docetaxel in 2D and 3D Models of Breast Cancer

    Background/aim: Breast cancer remains a major malignancy among women, and severe side effects and the development of acquired drug resistance frequently hinder current therapeutic strategies. The Notch signaling pathway, a key regulator of cell fate, is commonly dysregulated in breast cancer and associated with poor prognosis. Gamma-secretase inhibitors (GSIs) block Notch receptor activation and have shown potential anticancer efficacy. This study aimed to investigate the synergistic activity of two commonly used GSIs, DAPT and MK0752, combined with docetaxel or cisplatin in both 2D and 3D breast cancer models. Materials and methods: Triple-negative, highly metastatic MDA-MB-231 and ER+/PR+ MCF-7 breast cancer cell lines were treated with DAPT or MK0752 alone or in combination with docetaxel or cisplatin. Drug efficacy and potential synergism were evaluated in 2D monolayer cultures and 3D spheroid models. Sequential treatment strategies were also assessed, where docetaxel or cisplatin was administered prior to GSI exposure. Results: Both MDA-MB-231 and MCF-7 cell lines exhibited notable sensitivity to DAPT and MK0752 combinations with docetaxel or cisplatin in 2D and 3D cultures. Synergistic enhancement of cytotoxicity was observed, particularly in sequential treatment regimens. Pretreatment with docetaxel or cisplatin followed by GSI exposure demonstrated superior growth inhibition compared with either monotherapy or simultaneous combination treatments. Conclusion: This study highlights the therapeutic potential of combining GSIs with standard chemotherapeutics to overcome drug resistance in breast cancer. The observed synergy and sequencing effects provide a strong basis for further mechanistic and translational investigations to optimize GSI-based combinational therapy strategies.

    Brewing Property in Onedimensional City? Exploring Starbucks' Locational Strategies-Impacts in the Case of Izmir, Türkiye

    The article examines Starbucks'locational strategies and their impact on urban spaces, drawing on Rossi's concept of the "one-dimensional city." As global brands increasingly shape urban environments, three key effects emerge: rising property prices near stores, the concentration of locations in malls, transit hubs, and walkable areas, and alignment with urban landscapes standardized by major transportation infrastructure decisions. Similar to many cities in the Global South, Starbucks in & Idot;zmir functions both as a product and a driver of property-driven, rent-seeking urban development.Empirical evidence reveals that its locational strategies enhance accessibility, increase property values, and reshape the built environment. Through spatial analysis, the article examines Starbucks locations within a walkability and accessibility framework, highlighting their concentration in high-value districts with strong public transit connections.This research underscores how global brands reinforce socio-economic divides, transform urban spaces, and promote consumption-driven urbanization through their integration into global capitalism and real estate dynamics.

    State-Led Coastal Transformation in South Aegean Türkiye: Multiscalar Interventions and Oppositional Movements in the Case of Datça Muğla

    The article aims to reveal the contentious politics behind the state-led coastal transformation in South Aegean Türkiye by concentrating on the emblematic case of Datça Muğla. The research on Türkiye’s coastal transformation is still inadequate, particularly to analyze the role of the state, its policies, interventions and counter movements. The article underlies on the main argument that the coastal transformation in Türkiye manifests remarkable urban development to explore the role of the state policies and the socio-political reactions against them. The state initiated the coastal transformation process via the enforcement of key legislations and associated governmental regulations, and the top-down implementation of development plans and flagships projects. Fostered by the powerful market forces, these state-led practices are confronted with conflicts and social struggles at multiple scales of urban policymaking. The article draws on comprehensive research, employed multiple methods including a content analysis of coastal legislations, a documentary analysis of the key coastal plans and projects and lastly, a critical examination of some remarkable lawsuits, news and new governmental regulations on the coasts of Datça peninsula. As the case of Datça illustrates, a new conservation legislation at the macro-scale, the new yacht port and tourism-induced coastal development projects at the meso-scale, and the occupation of the public beaches by many legally constructed ways at the micro-scale, have consistently reconfigured the multiscalar interventions in the coastal transformation processes.     “Lost” at the Crossroads: An Exploration of Human Trafficking

    Human trafficking impacts a growing population of people in today’s world. Due to mounting social, political, and economic problems and ongoing wars and conflicts, millions are now forced to leave their home countries and are trying to illegally immigrate to other countries in search of enhanced living conditions. That is why human trafficking urgently requires global attention. Various forms of human trafficking take place in the Global South. Türkiye, located at the crossroads of Europe and Asia and bordered by eight countries, plays a pivotal role in the recently escalating trafficking activities. As a well-established migration route and a strategic location, it is both a source and target country in human trafficking. This study aims at analyzing two award-winning Turkish movies, Seaburners and More, which depict vulnerability, exploitation, and struggle of the underrepresented minority, from the perspectives of children and women. The results, obtained through manifest and latent content analysis, will shed light upon this significant issue with the aim of contributing to the global fight against human trafficking. The study portrays vulnerabilities at the individual level and explores the potential social, political, and economic reasons and consequences of human trafficking through media works. © Journal of Underrepresented and Minority Progress.

    Mock Alexander Polynomials

    In this paper, we construct Mock Alexander polynomials for starred links and linkoids in surfaces. These polynomials are defined as state summations on link or linkoid diagrams that satisfy f = n, where f denotes the number of regions and n denotes the number of crossings of diagrams.     Participatory Action for and Through Ecological Integrity: Toroidality as a Circular Design Model

    The ecological crisis corrupts natural cycles, damages networks of organisms, and disrupts the ecosystem equilibrium. In response to the ecological crisis, the concept of circularity proposes reformative actions. However, these actions are insufficient to reverse the crisis and achieve ecological integrity because they are non-holistic and based on human concerns and priorities. A more inclusive and integrative mindset is necessary for better-functioning circular processes. In this regard, this research presents a theoretical framework for an alternative circular design model – Toroidality – which is driven by collective knowledge and participatory action. Toroidality functions through the integrative capacity of collective knowledge and collaboration among human and non-human stakeholders in the ecosystem. Based on the intertwined cycles of collective knowledge and collective value-creation, Toroidality becomes a self-feeding circular design model. Through its four-phased circular design processes, Toroidality claims to generate solutions that have regenerative capacities. This article clarifies the conceptual background, theoretical framework and complementary notions to present the fundamentals of Toroidality. After this initial explanatory article, the dynamics of the phases and interactions among the stakeholders are aimed to be explored and reported in the following research, through a real-life case study. © 2025 Published by ITB Institute for Research and Community Services.

    Redundancy resolution options for the twin-it-romans robotic hybrid manufacturing system

    The Twinnig Iztech in Robotics Manufacturing System (TWIN-IT-ROMANS) project funded by EU Horizon -Widera-2023-Access-02-01 aims to develop a hybrid manufacturing system that can perform additive and subtractive manufacturing processes and inline quality control using a robotic system. The system will incorporate a 6-degree-of-freedom robot arm and a positioner with 2-degree-of-freedom, which will operate synchronously. This manipulation system is to be designed for performing different manufacturing operations with different degrees-of-freedom requirements. In order to reveal alternative trajectory planning scenarios for this system, this paper presents an initial review of redundancy resolution approaches for kinematically redundant robotic manipulators. First, the four main approaches for redundancy resolution techniques are introduced. Then main studies on energy minimization and stiffness maximization for kinematically redundant robotic manipulators are reviewed. Similar or new approaches are planned to be generated and implemented for the redundant system for hybrid manufacturing. © The Author(s), under exclusive license to Springer Nature Switzerland AG 2025.

    Can Fish Kills in Izmir Bay Be Explained With Satellite Image Analysis

    Motivated by a significant environmental crisis that emerged, where large numbers of dead fish washed ashore in İzmir Bay in the summer of 2024, this study aims to analyze the spatial and temporal dynamics of water quality in the inner bay prior to this incidence. By calculating indices such as NDCI, SABI, and UWQV, and correlating them with climatic data (air temperature, wind speed and relative humidity), this research seeks to document the occurrence and drivers of algal blooms in the bay using Landsat 8 and Sentinel-2 satellite data from 2017 to 2025. This is the first comprehensive study conducted for İzmir Bay that investigates the relationships between water quality indices and climatic variables. It also incorporates aerial analysis of the inner bay to provide a broader spatial perspective. A customized code using Python is developed for this study to independently download and analyze raw satellite data with respect to defined corrections/masks. The results of eight years of analysis indicated that critical conditions arise every summer with air temperatures reaching 40 degrees in the study area. Estimated aerial averaged NDCI index and Chl-a concentration values show a strong positive correlation with air temperature, particularly in the Spearman’s rank correlation (rs = 0.67 and 0.62 respectively), indicating a significant relationship between these parameters. Aerial distribution of the indices for the selected critical dates also revealed a significant increase in estimated Chl-a levels during the summer months, specifically in the regions determined from the risk maps produced as a result of this study. The areas with the greatest vulnerability coincide where Poligon, Ilıca streams in the south and Bostanlı and Çiğli streams in the north discharging into the bay. It is recommended that any planned external intervention methods for managing algal blooms should start with these highly vulnerable areas as presented by this study.

    Towards Sustainable Manufacturing: a Review and Future Directions in Additive Manufacturing of Fiber-Reinforced Polymer Composites

    The United Nations Sustainable Development Goals (SDGs) provide a global framework for addressing critical challenges such as climate change, resource scarcity and sustainable industrialization. With increasing demand for products and improving quality of life, linear consumption of materials and resources following the “take-make-waste” is no longer possible. As such, innovative solutions are increasingly necessary to enable circular economy in manufacturing. Additive manufacturing (AM) has emerged as a transformative technology in achieving SDGs by enhancing resource efficiency and minimizing waste. Fiber reinforced composites are a promising application of AM, as they offer the potential to optimize material use, reduce labor and support sustainable production practices. However, there is an urgent need for considering circular economy strategies, life cycle assessment (LCA) frameworks and effective recycling at the end of their lifetime. This study examines additive manufacturing systems for fiber-reinforced composites, their environmental impact and exploring the potential contributions of robotic integration in composite manufacturing to enhanced sustainability. © The Author(s), under exclusive license to Springer Nature Switzerland AG 2025.

    21st Century Urban Aesthetics in the Post Critical Age: Sanart Publication

    This research problematized the relationship between human and the built environment in the 21st century, which it describes as post-critical, and examined the relationship between them in a theoretical framework through aesthetics. Within this framework, it has analyzed selected articles from the proceedings book published by SANART (Association of Aesthetics and Visual Arts). This study has defined aesthetics as a way of relating human beings to their environment. It redefined holistic aesthetic thought through approaches that focus on uncertain, porous, relational boundaries and social processes. With new perspectives that reject the dualism of subject and object, the process associated with the concepts of movement and becoming have been conceptualized through the process of urban experience from the perspective of Manuel DeLanda. This study has analyzed the urban discourses presented in SANART publications with a focus on ’movement’. It has interpreted these movements as a feature of the built environment as it is shaped by the capacities of bodies and space. It has argued that a movementoriented evaluation of urban aesthetics would further the development of the critical field. © 2025, Istanbul Teknik Universitesi, Faculty of Architecture.
