    Tomato: Genetics, Genomics, and Breeding of Health-Related Traits

    Tomato (Solanum lycopersicum) is a popular crop due to its versatility and nutritional quality. In addition to its nutritional content, tomato is rich in various phytochemicals that are known to have beneficial effects on human health. These bioactive components include pigments like lycopene and Β-carotene, ascorbic acid (vitamin C), phenolic compounds, polyamines, and glycoalkaloids. Tomato metabolites have various bioactivities such as antihypertensive, antioxidant, anticancer, anti-inflammatory, antidiabetic, antiallergenic, antiatherogenic, antithrombotic, and antimicrobial effects. Research aimed at improving tomato for many of these specific activities is still in its infancy; however, a foundation of knowledge has been established for health-related (HR) traits in the crop. In this chapter, previous works surveying tomato germplasm for HR traits, conventional breeding, and genetic investigation of these characteristics are described. We also discuss research aimed at HR gene mapping and isolation as well as efforts to improve these traits via genetic engineering and genome editing. © Springer Nature Singapore Pte Ltd. 2023.

    Enhancing Wound Regeneration Potential of Fibroblasts Using Ascorbic Acid-Loaded Decellularized Baby Spinach Leaves

    Decellularization of plant tissues is an emerging route to fabricate scaffolds for tissue engineering and regenerative medicine. Although significant progress has been made in the field of plant tissue decellularization, functionalization of plant scaffolds is still an emerging field, and loading them with L-ascorbic acid to promote skin regeneration has not yet been reported. L-ascorbic acid is an antioxidant that plays a key role in collagen synthesis as a cofactor of lysyl hydroxylase and prolyl hydroxylase. It has been shown to have significant importance in physiological wound healing by stimulating fibroblasts to produce collagen at both the molecular and the genetic levels. In this work, we aimed to fabricate an ascorbic acid-releasing bioactive scaffold by introducing a stable form of ascorbic acid, L-ascorbic acid 2-phosphate (AA2P), into decellularized baby spinach leaves and investigated its biological activity in vitro. Our results demonstrated that AA2P could be easily introduced into decellularized baby spinach leaf scaffolds and subsequently released within the effective dose range. AA2P-releasing baby spinach leaves were found to increase metabolic activity and enhance collagen synthesis in L929 fibroblasts after 21 days. In conclusion, this study demonstrated the fabrication of a novel functionalized skin tissue engineering scaffold and made a significant contribution to the fields of plant decellularization and skin tissue engineering.

    A Screen-Printed Electrode Modified With Gold Nanoparticles/ Cellulose Nanocrystals for Electrochemical Detection of 4,4'-methylene Diphenyl Diamine

    Developing simple, cost-effective, easy-to-use, and reliable analytical devices if of utmost importance for the food industry for rapid in-line checks of their products that must comply with the provisions set by the current legislation. The purpose of this study was to develop a new electrochemical sensor for the food packaging sector. More specifically, we propose a screen -printed electrode (SPE) modified with cellulose nanocrystals (CNCs) and gold nanoparticles (AuNPs) for the quantification of 4,4'-methylene diphenyl diamine (MDA), which is one of the most important PAAs that can transfer from food packaging materials into food stuffs. The electrochemical performance of the proposed sensor (AuNPs/CNCs/SPE) in the presence of 4,4'- MDA was evaluated using cyclic voltammetry (CV). The modified AuNPs/CNCs/SPE showed the highest sensitivity for 4,4'-MDA detection, with a peak current of 9.81 mu A compared with 7.08 mu A for the bare SPE. The highest sensitivity for 4,4'-MDA oxidation was observed at pH = 7, whereas the detection limit was found at 57 nM and the current response of 4,4'-MDA rose linearly as its concentration increased from 0.12 mu M to 100 mu M. Experiments using real packaging materials revealed that employing nanoparticles dramatically improved both the sensitivity and the selectivity of the sensor, which can be thus considered as a new analytical tool for quick, simple, and accurate measurement of 4,4 '-MDA during converting operations.

    Fabrication and Characterization of a Solution Processed Flexible Thermal Sensor by Using Chemically Synthesized Go and Rgo

    Graphene oxide (GO) was reduced by ascorbic acid which is an environmental-friendly reductant and obtained sample was named as reduced GO (rGO). Stable dispersions of GO and rGO were prepared in N,N-Dimethylformamide (DMF). Compared to GO sample, rGO was determined to have more thermal stability, smaller sheet size and lower surface energy. GO and rGO dispersions were drop-casted on aluminum (Al) coated acetate substrate and used as thermal sensor. Fabricated sensors were tested from 25 °C to 150 °C. The sensors fabricated with GO, were not stabile against driven temperature changes. However, rGO ones, presented no thermal hysteresis effect after the first heating step. This sensor (Al/rGO/Al) acted like an NTC (Negative Temperature Coefficient) thermistor. The resistance of the rGO sensor was changed between 42 k? to 25 k? depending on the test temperature range (25 °C to 150 °C). Average beta value was calculated as 519.7649 K. © 2019 IEEE.

    Water-Soluble Antioxidant Potential of Turkish Pepper Cultivars

    In this work, 29 pepper cultivars that represent the diversity of types and varieties grown in Turkey were analyzed for water-soluble antioxidant capacity and phenolic and vitamin C contents. In addition, 14 non-Turkish cultivars were tested for comparison. Significant diversity was observed in the different cultivars with the most variation (7.4-fold) seen for total antioxidant capacity, which ranged from 2.57 to 18.96 mmol Trolox/kg. Vitamin C content for the peppers ranged from 522 to 1631 mg·kg-1, a 3.1-fold difference, whereas total phenolic content for the pepper cultivars ranged from 607 to 2724 mg·kg-1, a 4.5-fold difference. When cultivars were grouped by morphology/ use, it was found that some types had significantly more variation and higher antioxidant activities than other types. Thus, for water-soluble antioxidant capacity, most variation was seen in long, blunt-ended Çarliston types, whereas long, pointed Sivri peppers had the highest mean capacity. Bell-shaped Dolmalik and Sivri peppers had the most variation for phenolic content, but fancy Süs and Sivri types had the highest means for this trait. Dolmalik types showed the most variation for vitamin C content, whereas Süs and Sivri peppers had the highest means for this character. All three parameters were significantly and positively correlated with the strongest correlation between total antioxidant capacity and phenolic content (r = 0.71). The presence of significant variation for antioxidant content in Turkish germplasm indicates that this material can be used for improvement and genetic mapping of nutritional content in pepper.

    Structural Alteration of Cofactor Specificity in Corynebacterium 2,5-Diketo Acid Reductase

    Carynebacterium 2,5-Diketo-D-gluconic acid reductase (2,5-DKGR) catalyzes the reduction of 2,5-diketo-D-gluconic acid (2,5-DKG) to 2-Keto-L-gulonic acid (2-KLG). 2-KLG is an immediate precursor to L-ascorbic acid (vitamin C), and 2,5-DKGR is, therefore, an important enzyme in a novel industrial method for the production of vitamin C. 2,5-DKGR, as with most other members of the aldo-keto reductase (AKR) superfamily, exhibits a preference for NADPH compared to NADH as a cofactor in the stereo-specific reduction of substrate. The application of 2,5-DKGR in the industrial production of vitamin C would be greatly enhanced if NADH could be efficiently utilized as a cofactor. A mutant form of 2,5-DKGR has previously been identified that exhibits two orders of magnitude higher activity with NADH in comparison to the wild-type enzyme, while retaining a high level of activity with NADPH. We report here an X-ray crystal structure of the holo form of this mutant in complex with NADH cofactor, as well as thermodynamic stability data. By comparing the results to our previously reported X-ray structure of the holo form of wild-type 2,5-DKGR in complex with NADPH, the structural basis of the differential NAD(P)H selectivity of wild-type and mutant 2,5-DKGR enzymes has been identified.
