    Sensitive and Specific Detection of Ligands Using Engineered Riboswitches

    Riboswitches are RNA elements found in non-coding regions of messenger RNAs that regulate gene expression through a ligand-triggered conformational change. Riboswitches typically bind tightly and specifically to their ligands, so they have the potential to serve as highly effective sensors in vitro. In B. subtilis and other gram-positive bacteria, purine nucleotide synthesis is regulated by riboswitches that bind to guanine. We modified the xpt-pbuX guanine riboswitch for use in a fluorescence quenching assay that allowed us to specifically detect and quantify guanine in vitro. Using this assay, we reproducibly detected as little as 5 nM guanine. We then produced sensors for 2′-deoxyguanosine and cyclic diguanylate (c-diGMP) by appending the P1 stem of the guanine riboswitch to the ligand-binding domains of a 2′-deoxyguanosine riboswitch and a c-diGMP riboswitch. These hybrid sensors could detect 15 nM 2′-deoxyguanosine and 3 nM c-diGMP, respectively. Each sensor retained the ligand specificity of its corresponding natural riboswitch. In order to extend the utility of our approach, we developed a strategy for the in vitro selection of sensors with novel ligand specificity. Here we report a proof-of-principle experiment that demonstrated the feasibility of our selection strategy.

    Synthesis and Characterization of Two Oxo-Bridged Oxo(arylimido) [tris(3,5-Dimethylpyrazolyi)borato]molybdenum(v) Complexes and Crystal Structure of [motp*(o)ci](µ-O)[motp*(ci)(?nc6h 4ome)]

    Reaction of the oxo-molybdenum(V) compound, [MoTp*(O)Cl2], [Tp* = hydrotris(3,5-dimethylpyrazol-1-yl)borate] with p-methoxy and p-nitroaniline in the presence of Et3N under N2, afforded the oxo-bridged oxo(arylimido) molybdenum(V) complexes, [MoTp*(O)Cl](μ- O)[MoTp*(Cl)(≡NC6H4R)] (1, R = OMe; 2, R = NO2). The new compounds were characterized by elemental analysis, i.r., mass, and 1H-n.m.r. spectra. The single crystal X-ray crystallographic determination of [MoTp*(O)Cl](μ-O)[MoTp*(Cl) (≡NC6H4OMe)] was carried out to confirm that there is a Mo - O - Mo bridge and a near linear arylimido group in the structure.

    Synthesis and Characterization of a New Mono-Molybdenated Metallo-Ligand [mo{hb(3,5-Me2c3hn2)3}(no)cl(oc 6h4pph2-P)]

    The "metallo-ligands" can be considered effectively as a modified triphenylphosphine ligand, and used to generate novel transtion metal complexes in which electron-transfer and magnetism is embedded within the stable ligand framework. A new mono-molybdenated metallo-ligand was synthesized and characterized by NMR (1H and 31P-{1H}), IR and mass spectroscopy techniques as a starting material in the search for designing novel bimetallic complexes exhibiting optoelectronic properties and development of non-linear optical materials that possess commercial device applications.
