    A Comparative Study of Attention-Augmented YOLO Architectures for Defect Detection in Fused Deposition Modelling

    Additive manufacturing (AM), particularly fused deposition modelling (FDM), facilitates the fabrication of complex geometries with increasing flexibility and efficiency. Ensuring consistent print quality in FDM processes necessitates the development of accurate defect detection mechanisms. Attention-augmented YOLO (You Only Look Once) models have emerged as a promising solution for addressing this challenge. In this study, we systematically benchmark and evaluate the performance of YOLO architectures enhanced with attention mechanisms within the context of FDM 3D printing applications. The models were trained and evaluated using representative defect datasets. The attention-augmented models demonstrate improved detection performance. © 2025 IEEE.

    Çelik Fiber Katkısının Farklı Boyuna Donatı Oranına Sahip Betonarme Döşemelerin Zımbalama Davranışı Üzerinde Etkileri

    Sunulan çalışmada her iki yönde birbirine dik 0,004 (D1 serisi) ve 0,002 (D2 serisi) oranında boyuna donatıiçeren 2150x2150x150 mm boyutlarında iki grup betonarme döşeme, hacimce %0, %0,5, %1 ve %1,5oranında çelik fiber katkısı içeren beton karışımlarıyla dökülmüştür. Üretilen toplam sekiz döşeme ortanoktalarından statik yük altında test edilmişlerdir. Çelik fiber katkısı olmayan numunelerde yüksek boyunadonatı oranına sahip döşeme boyuna donatısında akma gerçekleşmeden gevrek bir şekilde zımbalamagöçmesi oluşurken düşük boyuna donatı oranına sahip döşeme zımbalama gerçekleşmeden önce çok dahasünek bir davranış göstermiştir. Çelik fiber katkısı her iki boyuna donatı oranında da iki kata varan oranlardazımbalama dayanımı artışlarına sebep olmuştur. Ancak D1 serisi döşemelerde çelik fiber katkısı maksimumyer değiştirmeleri önemli ölçüde arttırırken D2 serisinde maksimum yer değiştirmelerde önemli bir farkoluşmamış, bu döşemelerin yer değiştirmesi boyuna donatının akması tarafından kontrol edilmiştir. Çelikfiber katkısı oranının arttırılması D1 serisi döşemelerde dayanımın ve maksimum yer değiştirmelerinartmasına sebep olurken, D2 serisi döşemelerde %1'in üstü çelik fiber katkı oranları davranışta önemli birfark oluşturmamıştır. Yapılan deneyler Kritik Kesme Çatlağı Teorisi kullanılarak analitik olarakmodellenmiş ve bu tip modelleme ile ilgili bazı iyileştirmeler önerilmiştir.

    A Capsular Polysaccharide from a Healthy Human Microbiota Member Activates a Lag-3-NK Cell Axis to Restrain Colon Cancer and Augment Immunotherapy

    Colorectal cancer (CRC) is increasing globally, making identification of preventative measures necessary. Transplantation of the microbiota from CRC and non-CRC patients into mice demonstrates that non-diseased individuals possess organisms that reduce tumor formation and highlights Bacteriodes uniformis as protective. B. uniformis is reduced in humans with CRC, and proactive treatment with B. uniformis slows tumor growth in mice. Natural killer (NK) cells, but not T cells, are required for B. uniformis-mediated protection. CRC is recalcitrant to immunotherapies; however, addition of B. uniformis restores response to alpha-CTLA-4 treatment in an NK cell-dependent manner. We report that high Lag-3 expression is associated with greater survival in CRC patients and that B. uniformis-mediated protection is reliant on Lag-3 in innate cells. Induction of NK cell activity and reduced tumor growth is dependent on a specific B. uniformis capsular polysaccharide. Thus, healthy individuals possess tumor suppressor microbes that prevent cancer development and can be harnessed therapeutically.

    Dysfunctional K+ Homeostasis as a Driver for Brain Inflammation

    The central nervous system (CNS) relies on precise regulation of potassium ion (K+) concentrations to maintain physiology. This regulation involves complex cellular and molecular mechanisms that work in concert to regulate both intracellular and extracellular K+ levels. Inflammation, a key physiological response, encompasses a series of cell-specific events leading to inflammasome activation. Perturbations in K+-sensitive processes can result in either chronic or uncontrolled inflammation, highlighting the intricate relationship between K+ homeostasis and inflammatory signalling. This review explores molecular targets that influence K+ homeostasis and have been implicated in inflammatory cascades, offering potential therapeutic avenues for managing inflammation. We examine both cell-specific and common molecular targets across different cell types, providing a comprehensive overview of the interplay between K+ regulation and inflammation in the CNS. By elucidating these mechanisms, we identify leads for drug discovery programmes aimed at modulating inflammatory responses. Additionally, we highlight potential consequences of targeting individual molecular entities for therapeutic purposes, emphasizing the need for a nuanced approach in developing anti-inflammatory strategies. This review considers current knowledge on K+-sensitive inflammatory processes within the CNS, offering critical insights into the molecular underpinnings of inflammation and potential therapeutic interventions. Our findings underscore the importance of considering K+ homeostasis in the development of targeted therapies for inflammatory conditions within the CNS. © 2025 Elsevier B.V., All rights reserved.

    Redundancy resolution options for the twin-it-romans robotic hybrid manufacturing system

    The Twinnig Iztech in Robotics Manufacturing System (TWIN-IT-ROMANS) project funded by EU Horizon -Widera-2023-Access-02-01 aims to develop a hybrid manufacturing system that can perform additive and subtractive manufacturing processes and inline quality control using a robotic system. The system will incorporate a 6-degree-of-freedom robot arm and a positioner with 2-degree-of-freedom, which will operate synchronously. This manipulation system is to be designed for performing different manufacturing operations with different degrees-of-freedom requirements. In order to reveal alternative trajectory planning scenarios for this system, this paper presents an initial review of redundancy resolution approaches for kinematically redundant robotic manipulators. First, the four main approaches for redundancy resolution techniques are introduced. Then main studies on energy minimization and stiffness maximization for kinematically redundant robotic manipulators are reviewed. Similar or new approaches are planned to be generated and implemented for the redundant system for hybrid manufacturing. © The Author(s), under exclusive license to Springer Nature Switzerland AG 2025.

    Towards Sustainable Manufacturing: a Review and Future Directions in Additive Manufacturing of Fiber-Reinforced Polymer Composites

    The United Nations Sustainable Development Goals (SDGs) provide a global framework for addressing critical challenges such as climate change, resource scarcity and sustainable industrialization. With increasing demand for products and improving quality of life, linear consumption of materials and resources following the “take-make-waste” is no longer possible. As such, innovative solutions are increasingly necessary to enable circular economy in manufacturing. Additive manufacturing (AM) has emerged as a transformative technology in achieving SDGs by enhancing resource efficiency and minimizing waste. Fiber reinforced composites are a promising application of AM, as they offer the potential to optimize material use, reduce labor and support sustainable production practices. However, there is an urgent need for considering circular economy strategies, life cycle assessment (LCA) frameworks and effective recycling at the end of their lifetime. This study examines additive manufacturing systems for fiber-reinforced composites, their environmental impact and exploring the potential contributions of robotic integration in composite manufacturing to enhanced sustainability. © The Author(s), under exclusive license to Springer Nature Switzerland AG 2025.

    Search for Supersymmetry in pp Collisions at √s=7 TeV in Events With a Single Lepton, Jets, and Missing Transverse Momentum

    Results are reported from a search for new physics processes in events containing a single isolated high-transverse-momentum lepton (electron or muon), energetic jets, and large missing transverse momentum. The analysis is based on a 4.98 fb(-1) sample of proton-proton collisions at a center-of-mass energy of 7 TeV, obtained with the CMS detector at the LHC. Three separate background estimation methods, each relying primarily on control samples in the data, are applied to a range of signal regions, providing complementary approaches for estimating the background yields. The observed yields are consistent with the predicted standard model backgrounds. The results are interpreted in terms of limits on the parameter space for the constrained minimal supersymmetric extension of the standard model, as well as on cross sections for simplified models, which provide a generic description of the production and decay of new particles in specific, topology based final states.

    Role of Long Non-Coding RNA X-Inactive Transcript (XIST) in Neuroinflammation and Myelination: Insights From Cerebral Organoids and Implications for Multiple Sclerosis

    Background/Objectives: X-inactive-specific transcript (XIST) is a factor that plays a role in neuroinflammation. This study investigated the role of XIST in neuronal development, neuroinflammation, myelination, and therapeutic responses within cerebral organoids in the context of Multiple Sclerosis (MS) pathogenesis. Methods: Human cerebral organoids with oligodendrocytes were produced from XIST-silenced H9 cells, and the mature organoids were subsequently treated with either FTY720 or DMF. Gene expression related to inflammation and myelination was subsequently analyzed via qRT-PCR. Immunofluorescence staining was used to assess the expression of proteins related to inflammation, myelination, and neuronal differentiation. Alpha-synuclein protein levels were also checked via ELISA. Finally, transcriptome analysis was conducted on the organoid samples. Results: XIST-silenced organoids presented a 2-fold increase in the expression of neuronal stem cells, excitatory neurons, microglia, and mature oligodendrocyte markers. In addition, XIST silencing increased IL-10 mRNA expression by 2-fold and MBP and PLP1 expression by 2.3- and 0.6-fold, respectively. Although XIST silencing tripled IBA1 protein expression, it did not affect organoid MBP expression. FTY720, but not DMF, distinguished MBP and IBA1 expression in XIST-silenced organoids. Furthermore, XIST silencing reduced the concentration of alpha-synuclein from 300 to 100 pg/mL, confirming its anti-inflammatory role. Transcriptomic and gene enrichment analyses revealed that the differentially expressed genes are involved in neural development and immune processes, suggesting the role of XIST in neuroinflammation. The silencing of XIST modified the expression of genes associated with inflammation, myelination, and neuronal growth in cerebral organoids, indicating a potential involvement in the pathogenesis of MS. Conclusions: XIST may contribute to the MS pathogenesis as well as neuroinflammatory diseases such as and Alzheimer's and Parkinson's diseases and may be a promising therapeutic target.

    Plasma Proteomic Markers of Interleukin-1β Pathway Associated With Incident Age-Related Macular Degeneration in Persons With Aids

    Objective To evaluate the associations of plasma inflammatory proteins with age-related macular degeneration (AMD) in persons with the AIDS, using a discovery-based proteomics approach. Design A nested case-control study (analysis 1) and nested cohort study (analysis 2). Participants Persons with AIDS enrolled in the Longitudinal Study of the Ocular Complications with AIDS (LSOCA). Methods Cryopreserved plasma specimens obtained at baseline were assayed for inflammatory proteins using the Olink Inflammation Explore Panel 1. In analysis 1, baseline proteomic profiles for 26 persons with AIDS and incident intermediate-stage AMD 5 to 10 years after baseline and 49 matched controls (matched for age, biologic sex, race/ethnicity, and follow-up) without AMD were compared. In analysis 2, 475 persons from LSOCA with baseline plasma inflammatory proteomic profile measurements were followed for incident cataract and mortality. Main Outcome Measures Incident intermediate-stage AMD; incident cataract; and mortality. Results Of 365 measurable plasma inflammatory proteins, 118 (32%) were associated with incident intermediate-stage AMD at the false discovery rate-adjusted Q < 0.05 level after adjustment for smoking, CD4+ T count, and plasma human immunodeficiency virus RNA level. Gene ontology pathway enrichment analysis identified the interleukin (IL)-1 beta pathway and wound healing pathways, including tissue inhibitor of metalloproteinase 3, as significantly associated with incident AMD. These associations were qualitatively different from those associated with incident cataracts, where elevated levels of inflammatory proteins were associated with a decreased risk of cataracts. A much broader number of inflammatory pathways, including those related to the adaptive immune system, were associated with mortality. Conclusions Upregulation of the IL-1 beta pathway appears to be associated with an increased risk of incident AMD in persons with AIDS. Given the availability of inhibitors of this pathway, inhibition of the IL-1 beta pathway may provide a therapeutic avenue for treatment of AMD.     A Review of the Experimental and Numerical Studies on the Compression Behavior of the Additively Produced Metallic Lattice Structures at High and Low Strain Rates

    Recent advances in additive manufacturing have enabled the construction of metallic lattice structures with tailored mechanical and functional properties. One potential application of metallic lattice structures is in the impact load mitigation where an external kinetic energy is absorbed by the deformation/ crushing of lattice cells. This has motivated a growing number of experimental and numerical studies, recently, on the crushing behavior of additively produced lattice structures. The present study overviews the dynamic and quasi-static crushing behavior of additively produced Ti64, 316L, and AlSiMg alloy lattice structures. The first part of the study summarizes the main features of two most commonly used additive processing techniques for lattice structures, namely selective-laser-melt (SLM) and electrobeam-melt (EBM), along with a description of commonly observed process induced defects. In the second part, the deformation and strain rate sensitivities of the selected alloy lattices are outlined together with the most widely used dynamic test methods, followed by a part on the observed microstructures of the SLM and EBM-processed Ti64, 316L and AlSiMg alloys. Finally, the experimental and numerical studies on the quasi-static and dynamic compression behavior of the additively processed Ti6 4, 316L, and AlSiMg alloy lattices are reviewed. The results of the experimental and numerical studies of the dynamic properties of various types of lattices, including graded, non-uniform strut size, hollow, non-uniform cell size, and bio-inspired, were tabulated together with the used dynamic testing methods. The dynamic tests have been noted to be mostly conducted in compression Split Hopkinson Pressure Bar (SHPB) or Taylor-and direct-impact tests using the SHPB set-up, in all of which relatively small-size test specimens were tested. The test specimen size effect on the compression behavior of the lattices was further emphasized. It has also been shown that the lattices of Ti6 4 and AlSiMg alloys are relatively brittle as compared with the lattices of 316L alloy. Finally, the challenges associated with modelling lattice structures were explained and the micro tension tests and multi-scale modeling techniques combining microstructural characteristics with macroscopic lattice dynamics were recommended to improve the accuracy of the numerical simulations of the dynamic compression deformations of metallic lattice structures.
