    Trna Wobble Base Modifications and Boric Acid Resistance in Yeast: Boron-Resistant Deletion Mutants Induce the General Amino Acid Control Mechanism and Activate Boron Efflux

    Boric acid is essential for plants and has many vital roles in animals and microorganisms. However, its high doses are toxic to all organisms. We previously screened yeast deletion collections to identify boric acid-resistant and susceptible mutants to identify genes that play a role in boron tolerance. Here, we analyzed boron resistant mutants (elplΔ, elp3Δ, elp6Δ, ncs2Δ, ncs6Δ and ktil2Δ) for their abilities to modulate the general amino acid control system (GAAC) and to induce boron efflux pump ATR1. The mutants analyzed in this study lack the genes that play roles in tRNA Wobble base modifications. We found that all of the boron resistant mutants activated Gcn4-dependent reporter gene activity and increased the transcript level of the ATR1 gene. Additionally, boron resistant cells accumulated less boric acid in their cytoplasm compared to the wild type cells upon boron exposure. Thus, our findings suggested that loss of wobble base modifications in tRNA leads to GAAC activation and ATR1 induction, which in turn reduced intracellular boron levels and caused boron resistance.

    Structural Performance of R/C Buildings in 2023 Kahramanmaraş Earthquakes Under the Lens of Hassan Index

    Starting with the 1999 Kocaeli Earthquake, seismic damage indices have been developed to quantify damage potential. The late Professor Mete Sozen played a significant role in this endeavor by introducing the Hassan Index, which is an indirect measure of the robustness of structures. Its calculation is straightforward, given the vertical structural element dimensions at ground level. Various earthquakes worldwide, from Taiwan to Chile, have provided opportunities to assess its effectiveness across diverse building practices and earthquake characteristics. Following the February 2023 Kahramanmaras Earthquakes, a reconnaissance team was dispatched to gather field data to observe the damages through the lens of the Hassan Index. The study extended to cover all major city and town centers, studying around 250 reinforced concrete buildings with 2-16 stories. These buildings were all constructed after the 2000s. Hence, the data set sheds light on the performance of Turkish construction practices after the commencement of the 1998 Turkish Building code and the following earthquake regulations. By evaluating the newly gathered data alongside the existing dataset, a basis is established to consider the Hassan Index as a preliminary design index rather than solely as a seismic damage index.

    Pedestrian Equipment Anomaly Detection With Computer Vision in Warehouses

    The rapid growth of the logistics sector in recent years caused the expansion of warehouse areas and the increase in the number of equipment used. With the increase in these activities, the possibility of work accidents in warehouses also increases. In defiance of this situation, it has been determined that a real-time prediction system of pedestrian and equipment interaction is needed to ensure in-warehouse reliability. This system should address the urgent need to reduce the risk of work accidents and focus on the overall goal of reducing the possibility of work accidents in warehouse environments. To overcome this challenge, we propose a comprehensive Warehouse Anomaly Detection and Control System consisting of object detection, object tracking, action detection, and alarm classification components which will play an important role in increasing work safety in warehouse environments. YOLOv7 (You Only Look Once version 7) is a deep learning model that detects objects quickly and accurately in a single network pass. The deep learning-based Deep SORT algorithm used for object tracking provides a dynamic understanding of the warehouse environment by continuously storing these identified problems in real-time. The action detection part of this system is designed to identify and analyze actions and movements, recognizing anomalies and potential risks. In this part, the speed of pedestrians and equipment are detected utilization of 3D bounding boxes of objects and perspective transformation. The possible accident risks are measured using the intersection percentage of these areas, the magnitude of speed, the direction of the motion vector of pedestrian and equipment, and the distances between objects. Alert levels can be considered as encounter, near-miss, and emergency. Using this system in warehouses will reduce the risk of possible work accidents that may even result in death. © 2024, Avestia Publishing. All rights reserved.

    Discussion on the Causes of the Observed Damages in the 2023 Kahramanmaraş Earthquakes

    The devastating earthquakes in February 2023 in south-east Turkiye demonstrated that the Turkish built environment does not meet the needed performance. The frequency of earthquakes in Turkiye calls for urgent organization to upgrade the existing structures and avoid the repetition of deficient applications. Hence, it is necessary to identify the causes of the damage. The presented study concentrated on a subset of the existing inventory built under the governance of 1998 and later Turkish Earthquake Codes. Following the earthquakes in Kahramanmaras, Turkiye, a reconnaissance study was conducted to understand the causes of the damage incurred. The extensive study covered all the major city and town centers, specifically concentrating on the 2- to 16-story reinforced concrete buildings. The first impression of the data gathered indicates significant detailing imperfections. Poor drift control, inferior layout preferences, and non-structural detailing issues are the other suspects. The observed damage and the current seismic regulation requirements will be critically discussed to form a basis for possible future improvements to the seismic code.

    The Statistical Assessment of Variables Affecting Conservation Condition for Historic Houses

    Conservation condition is the structural state of a historic building at a particular time. This study aims to determine the variables affecting conservation condition of heritage buildings in historic urban sites. The methodology includes selection of the case study, site survey and listing of historic house characteristics with conventional tools of architectural conservation, and determination of variables affecting their conservation condition with statistical tools. The houses in a portion of Kuyulu neighborhood, and in a portion of Kurtuluş Street, presenting variety in terms of built heritage characteristics in Antakya historic urban site, which experienced a destroying earthquake sequence in 2023, were focused on. The dataset prepared in 2019 site survey is examined by T-tests, ANOVA, regression, and exploratory spatial data analysis statistical tools. As a result, construction technique, land use, and number of storeys were determined as significant variables affecting conservation condition. While addressing abandonment issues and considering both commercial and residential functions for adaptive reuse can positively affect conservation conditions, it is crucial to recognize buildings with combined construction systems show a negative effect on conservation condition which should not be preferred in future constructions and need priority for consolidation interventions. Meanwhile, the construction period and alterations are revealed as insignificant variables on conservation condition. The study concludes that systematic planning, guided by statistical insights, can prioritize interventions and enhance positive variables corresponding to the heritage qualities that have positive impact on conservation condition while mitigating negative ones, thus ensuring the preservation of historic urban sites. © 2024, Avestia Publishing.     Peroxisome Proliferator-Activated Receptor Alpha (pparα) Rs4253778 Polymorphism in a Turkish Soccer Player Cohort

    Aim: Peroxisome proliferator-activated receptor alpha gene (PPARA) is one of the important gene determining athletes endurance capacity. The product of the gene functions in the regulation of lipid metabolism, in mostly heart and skeletal system, and maintaining glucose homeostasis and biogenesis in mitochondria. In this study, we aimed to determine the allelic distribution of PPARA rs4253778 polymorphism in a Turkish soccer player cohort. Methods: We enrolled 22 professional soccer players and 68 sedentary controls for the study. Buccal cells were used for DNA sources, and real- time PCR methodology was followed for genotyping. GG genotypes were superior in both groups, and this was statistically significant when we compare the genotypes between groups. Result:When we consider the alleles, G allele was found more frequently in players, and in controls. When we compare two groups, we determine a statistically significant difference, indicating the effect of G allele in soccer physiology. Conclusion: Our findings suggest that PPARA rs4253778 polymorphism is an effective genetic factor in soccer physiology, and should be considered as an important genetic biomarker in sports genetics. © 2020 Pamukkale University.     Evaluating Performance and Reliability of Selective Redundant Multithreading for Gpgpu Applications

    With the widespread use of GPU architectures in general-purpose computations, evaluating the soft error vulnerability of GPGPU programs and employing efficient fault tolerance techniques for more reliable execution becomes more prominent. Performing full redundancy, based on the redundant execution of the complete program, results in resource consumption and performance loss as well as energy inefficiency. Therefore, determining the most error-prone regions of the target program code and replicating only those parts maintains both high performance and acceptable error rates. In this study, we propose a partial redundant multithreading mechanism based on the soft error vulnerability of GPGPU applications and perform a trade-off analysis between performance and reliability. Firstly, we perform fault injection experiments to evaluate the SDC rates for each kernel function. Then, based on the outcome of the fault injection experiments, we determine the kernel function to-be-replicated. According to the pragmas denoting the redundancy points in the source code, our custom LLVM pass generates the code that enables the redundant execution for the specified code region. We evaluate both the reliability and performance of the redundant execution scenarios measuring the execution time of the redundant program generated by our compiler-managed redundancy technique. Our results demonstrate that protecting only the most vulnerable kernel functions enables high reliability without hurting the performance significantly. © 2021 The Authors.

    Artist Recommendation Based on Association Rule Mining and Community Detection

    Recent advances in the web have greatly increased the accessibility of music streaming platforms and the amount of consumable audio content. This has made automated recommendation systems a necessity for listeners and streaming platforms alike. Therefore, a wide variety of predictive models have been designed to identify related artists and music collections. In this paper, we proposed a graph-based approach that utilizes association rules extracted from Spotify playlists. We constructed several artist networks and identified related artist clusters using Louvain and Label Propagation community detection algorithms. We analyzed internal and external cluster agreements based on different validation criteria. As a result, we achieved up to 99.38% internal and 90.53% external agreements between our models and Spotify's related artist lists. These results show that integrating association rule mining concepts with graph databases can be a novel and effective way to design an artist recommendation system.     Entrepreneurial Networks in the Ottoman Empire: the Case of Osman Inayet

    This article analyzes the dynamics of an entrepreneurial activity, mine exploitation, at the end of the nineteenth century in Salonica through the economic activities of an entrepreneur, Osman Inayet and proposes an evaluation of the complex relationships he created with other entrepreneurs through mining activities. Legal transformations concerning mining activities coupled with the economic growth due to the commercial expansion especially in port-cities such as Salonica in this period paved the way for an increase in the interest in mining activities. The mining activities of Osman Inayet, one of the entrepreneurs engaged in this sector, serve to analyze the components of the economic network he formed around this activity and lead us to enrich our comprehension about the nature of economic relations in Salonica at the end of the nineteenth century. Focusing on an individual case provides a more comprehensive picture of individual choices, decisions, and experiences. Furthermore, this analysis, based on archival documents on mine concessions, provides new perspectives to our understanding of the inner mechanisms entrepreneurial activities. © 2020 by the authors.
