    Super Citizens: the Power of Social Innovation in İzmir/Turkey

    This study aims to define the characteristics of social innovators, measure the impact of social innovation, and investigate its effects on urban transformation in İzmir. The study is based on semi-structured interviews with 14 social innovators and conducted by adopting a conceptual framework. We identify the characteristics of ‘super citizens’ in the literature that can contribute to SI’s empowerment in cities. The first finding shows that these individuals are expected to have 8 characteristics: being an activist, a social entrepreneur, a mediator, having a wide network, organizing goal-oriented works, involving all actors in the process, producing new solutions focused on the subject and finally being able to keep up with the change. The second finding shows that they are the most important actors in sustaining urban development, and their projects have impacts on urban transformation from a city level to an international level. If the interaction mechanism between relevant actors is provided by the local government through the Penta-helix model, super citizens may become one of the primary stakeholders to promote SI and create a more sustainable urban development. © 2024 The European Association for the Advancement of the Social Sciences.

    Re-Introducing Statutory Regional Spatial Planning Strategies in England: Reflections Through the Lenses of Policy Integration

    Statutory regional spatial strategies were abolished in England, United Kingdom in 2010. There are, however, increasing calls in favour of a re-introduction of statutory comprehensive spatial strategies at the regional level to enhance integrated economic growth and address exacerbating spatial inequalities. Through a survey and in-depth interviews conducted with experts and policymakers of the North-West region of England, this paper explores whether the introduction of such statutory strategies could find justificative grounds through policy integration of transportation, housing, and employment policies. We conclude with a set of mechanisms that could foster this re-introduction serving regional geographies beyond the North-West.

    From Concrete To Abstract Regional Planning Strategies in North West England: Building and Legitimizing Discourses and Mobilizing Actors for Spatial Transformation?

    This study questions if abstract regional planning strategies are fit to respond to changing societal and political conditions. We compare regional planning strategy making in North West England. Findings suggest that abstract strategies are more effective in building than managing transformative discourses. Results show that: (I) transformative discourses need to be built around manageable regional socio-spatial and spatial-economic disparities; (II) policy entrepreneurs should be targeted with equal consideration for power and counterpower; and (III) the regional planning authority should have access to specific funding schemes. It is our ultimate aim to re-energize strategic regional planning debates in England and beyond. © 2021 Informa UK Limited, trading as Taylor & Francis Group.

    Therapeutic Potential of Apigenin, a Plant Flavonoid, for Imatinib-Sensitive and Resistant Chronic Myeloid Leukemia Cells

    Despite the presence of many therapeutic regimens like imatinib and other tyrosine kinase inhibitors, the development of resistance, intolerance, and side effects makes chronic myeloid leukemia (CML) therapy challenging. Thus, there is a need to discover novel drugs for CML patients. In this study, we attempted to assess apigenin, a common plant dietary flavonoid, in terms of its cytotoxic, apoptotic, and cytostatic effects on imatinib-sensitive and resistant Philadelphia-positive CML cells. We analyzed apigenin's effects on cell proliferation, apoptosis, caspase-3 activity, loss of mitochondrial membrane potential, and cell cycle progression in K562 and K562/IMA3 cells. Furthermore, we described genes and gene networks that are modulated in CML in response to apigenin. Results of our study revealed that apigenin has cytotoxic and apoptotic effects on both cell types. We also displayed that apigenin induced G2/M arrest in K562 cells while arresting K562/IMA3 cells in S phase especially at the highest apigenin concentration. The expression analysis identified a set of genes that were regulated by apigenin in K652 and K562/IMA3 cells. Association of modulated genes with biological functional groups identified several networks affected by apigenin including cell survival, proliferation, cell death, cell cycle, and cell signalling pathways.

    Resveratrol Triggers Apoptosis Through Regulating Ceramide Metabolizing Genes in Human K562 Chronic Myeloid Leukemia Cells

    Resveratrol, an important phytoalexin in many plants, has been reported to have cytotoxic effects on various types of cancer. Ceramide is a bioactive sphingolipid that regulates many signaling pathways, including cell growth and proliferation, senescence and quiescence, apoptosis, and cell cycle. Ceramides are generated by longevity assurance genes (LASS). Glucosylceramide synthase (GCS) and sphingosine kinase-1 (SK-1) enzymes can convert ceramides to antiapoptotic molecules, glucosylceramide, and sphingosine-1-phosphate, respectively. C8:ceramide, an important cell-permeable analogue of natural ceramides, increases intracellular ceramide levels significantly, while 1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP) and SK-1 inhibitor increase accumulation of ceramides by inhibiting GCS and SK-1, respectively. Chronic myelogenous leukemia (CML) is a hematological disorder resulting from generation of BCR/ABL oncogene. In this study, we examined the roles of ceramide metabolizing genes in resveratrol-induced apoptosis in K562 CML cells. There were synergistic cytotoxic and apoptotic effects of resveratrol with coadministration of C8:ceramide, PDMP, and SK-1 inhibitor. Interestingly, there were also significant increases in expression levels of LASS genes and decreases in expression levels of GCS and SK-1 in K562 cells in response to resveratrol. Our data, in total, showed for the first time that resveratrol might kill CML cells through increasing intracellular generation and accumulation of apoptotic ceramides.
