    PEG-peptide conjugate containing cathepsin B degradation unit as a doxorubicin carrier system

    A drug delivery system (DDS) containing a cathepsin B degradable sequence and pH-responsive histidines was prepared by methoxypolyethylene glycol and peptide conjugation. Doxorubicin was attached to the carrier system using amide linkage to give the final form of the DDS, denoted as mPEG-AT3-DOX. mPEG-AT3-DOX exhibited a bimodal size distribution at about 15 and 30 nm independent of pH, whereas the size of the control DDS containing no peptide sequence, mPEG-DOX, was measured as ∼ 15–20 nm. At the end of 72 h, % doxorubicin release from both of the DDSs was observed to be below 8.5 ± 3% in the absence of cathepsin B, and it increased to 17 ± 2% in the presence of cathepsin B for mPEG-AT3-DOX. Complete degradation of AT3 peptide within 3 h upon incubation with cathepsin B suggests that lower than expected doxorubicin release is likely due to the aggregation tendency of mPEG-AT3-DOX. Absolute IC50 values indicated that the cytotoxicity trend of the samples is in the order of free DOX ≥ mPEG-AT3-DOX >mPEG-DOX. Considering these results, PEG-peptide-doxorubicin conjugates can be promising candidates in cancer therapy if they are designed to have more pronounced pH-responsive behavior to increase the drug release rate.

    Anticorrosion Coating for Magnesium Alloys: Electrospun Superhydrophobic Polystyrene/Sio2 Composite Fibers

    Superhydrophobic nanocomposite coatings for magnesium surfaces with remarkable corrosion resistance were fabricated by electrospinning in the presence of fluorosilane-functionalized silica (SiO2) nanoparticles. The effects of surface-modified silica (mod-SiO2) nanoparticles on the superhydrophobicity and corrosion resistance of polystyrene (PS)/mod-SiO2 fiber coatings were evaluated. The incorporation of the SiO2 nanoparticles endows PS fibers with rough surfaces exhibiting a water contact angle (WCA) of 165◦. The surface wettability, corrosion resistance, and their relation to the inorganic content in the PS fibers and the contact angle of the composite coatings were explored. Analysis of the corrosion results confirmed that the PS/mod-SiO2 coating protected the Mg surface from corrosion. In addition, PS fibers containing mod-SiO2 nanoparticles showed improved hydrophobicity, and excellent corrosion resistance was achieved with PS fibers containing 4 wt% SiO2 nanoparticles.

    Detection of the Contamination Sources of Listeria Monocytogenes in Pickled White Cheese Production Process Line and Genotyping With the Pulsed-Field Gel Electrophoresis Method

    This study was conducted to determine the contamination sources, serotyping profiles, and antibiotic resistance patterns of Listeria monocytogenes isolated during the production of pickled white cheese. The genetic-relatedness of the isolates to EGD SLCC (5835) (1/2a, lineage II) and ATCC (13932) (4b, lineage I) reference strains was also determined with pulsed-field gel electrophoresis (PFGE) as a result of digestions with AscI and ApaI enzymes. Samples were collected from 16 different points in the production process of 4 different plants at 3 different times. Among the 192 samples examined, 17 (8.85%) were determined to be contaminated with Listeria spp. Three isolates (3.53%) obtained from raw milk, wall/ground, and press cases were identified as L. monocytogenes via the conventional culture method and confirmed by polymerase chain reaction. These isolates were found to belong to serotype 4b. According to antibiotic resistance testing against 10 antibiotics (ampicillin, gentamicin, erythromycin, tetracycline, chloramphenicol, cefalotin, streptomycin, vancomycin, penicillin, and sulfamethoxazole/trimethoprim), it was determined that isolates from raw milk and press cases were resistant to erythromycin. PPGE band patterns of the isolates displayed indistinguishable with AscI and 80%-94% homology with ApaI. The isolates were observed to display high homology to ATCC (13932) and lower homology to EGD SLCC (5835) obtained by both enzymes.

    Reactions of the Solvent-Stabilized Compound [moocl2(thf)2] With Aromatic Nitrogen Donor Ligands: Spectroscopic Characterization and Semiempirical Am1* Calculations

    Reaction of the solvent-stabilized dioxo-molybdenum(VI) compound [MoO2 Cl2 (THF)2]withP Ph3 yielded the oxomolybdenum(IV) species [MoOCl2 (THF)2](1) by the removal of one oxygen atom as PPh3 O. The complexes [MoOCl2 (=NC6 H4 CN)] 2,[MoOCl2 (CNC6 H4 OH)] 3,[MoOCl2 (=NC6 H4 NH2)] 4,and [MoOCl2 (=NC6 H4 OCH3)] 5, were synthesized by the reactions of [MoOCl2 (THF)2]with XC6 H4 Y(X =NH2, Y = CN; X= CN, Y = OH; X = Y = NH2;X=NH2,Y=OCH3). All the new compounds were characterized by elemental analyses, and FTIR, UV-Vis, and 1 H and 13 C-NMR spectra. Geometrical parameters and molecular orbital calculations showed that compounds 2-5 are stabilized by a charge transfer between the Mo center and the phenyl ring.

    Application of Experimental Design on Determination of Aluminum Content in Saline Samples by Adsorptive Cathodic Stripping Voltammetry

    Adsorptive cathodic stripping voltammetric determination of aluminum at ng mL-1 levels in salt samples based on the metal complexation with Calcon (1-(2-hydroxynaphthylazo)-2 naphthol-4-sulfonic acid) and the subsequent adsorptive deposition onto a hanging mercury drop electrode was studied. Central composite design was used as a design method. Several chemical and instrumental parameters (pH, ligand concentration, deposition time, deposition potential, and complexing time) were involved in the experimental design. Analytical parameters such as repeatability, linearity, and accuracy were also investigated and the detection limit was found as 0.32 ng mL-1.

    Hydrogenation of Citral Over Ni and Ni-Sn Catalysts

    Liquid phase citral hydrogenation over zeolite-supported monometallic Ni and bimetallic Ni-Sn catalysts was studied. The zeolite support materials were Na-Y, Na-mordenite, and clinoptilolite. Ni and Sn contents of the monometallic and bimetallic catalysts were 8.1-9.2 wt% and 0.46 wt%, respectively. The type of the zeolite support affected the activity and selectivity of the catalysts differently. The main product of the citral hydrogenation reaction was citronellal, for both monometallic (84.5% yield) and bimetallic (44.5% yield) catalysts. The addition of promoter increased the selectivity to unsaturated alcohols (geraniol+nerol), i.e. it changed from 0.9% to 6.3% over mordenite and from 0.9% to 2.1% over Na-Y-supported catalysts. Furthermore, activity of the Ni catalysts decreased while the quantity of acetal remained almost constant. Intimate contact between active metal, promoter, and support, and a catalyst with a high concentration of weak acid sites gave high selectivity to geraniol+nerol.

    Hydrogenation of Citral Over Pt and Pt-Sn Catalysts

    Hydrogenation of citral on monometallic (Pt) and bimetallic (Pt-Sn) catalysts was investigated. It was found that the catalyst activities and product distributions were different over Na-Y and Clino supports. Among monometallic catalysts, Pt/Na-Y was more selective to citronellol (3.9%) and unsaturated alcohols, geraniol and nerol, (14.1%). Sn addition increased the catalyst activities. However, its effect on product distribution differed. Yield of geraniol+nerol changed from 5.1% to 19.7% over Clino. However, selectivity to unsaturated alcohols over Na-Y decreased when it was prepared as a bimetallic catalyst support. Unsaturated alcohol formation was favored when there was a metal support interaction over monometallic catalysts, and metal-promoter interaction over bimetallic catalyst.

    Genetic Multivariate Calibration Methods for Near Infrared (nir) Spectroscopic Determination of Complex Mixtures

    The simultaneous determination of ternary mixtures of methylene chloride, ethyl acetate, and methanol using near infrared (NIR) spectroscopy and 4 different genetic algorithms based multivariate calibration methods was demonstrated. The 4 genetic multivariate calibration methods are genetic partial least squares (GPLS), genetic regression (GR), genetic classical least squares (GCLS) and genetic inverse least squares (GILS). The sample data set contains the NIR spectra of 63 ternary mixtures and covers the range from 900 to 2000 nm in 2 nm intervals. Of these 63 spectra, 42 were used as the calibration set, and 21 were reserved for the prediction purposes. Several calibration models were built with the 4 genetic algorithm based methods for each component that makes up the mixtures. Overall, the standard error of calibration (SEC) and the standard error of prediction (SEP) were in the range of 0.22 to 2.5 (% by volume (v/v)) for all the 4 methods. A comparison of genetic algorithm selected wavelengths for each component and for each method was also included.
