    Hdac9/P300 Immunoexpression and Migration Analysis for Malignant Melanoma Stem Cell

    Melanoma is an aggressive tumor with a poor prognosis that worsens in the metastatic phase. Distruptions of epigenetic mechanisms is known to effect cancer stem cells (CSCs) activity. Malignant melanoma (MM) progression may be promoted by changes in the genetic structure of CSC. Thus, treatments that target epigenetic modifications could be a promising weapon, especially in melanoma. Here, we compared p300, HDAC9, and Factin proteins in melanoma CSCs (CD133+), non-CSCs (CD133-) and CHL-1 cell line, as well as cell migration and division rates. At 4 and 6 h, P300 protein levels in CHL-1 and CD133 + were remarkably similar, and the CD133- showed increases in expression levels as the incubation period lengthened. HDAC9 protein intensity decreased in CHL-1, increased in the CD133-, and remained relatively unchanged in the CD133+ as the incubation period lengthened. The mean value of F-actin expression level increased in all cell group with time, when the highest increase observed in CHL-1. In conclusion, our studies contribute to the management of metastatic diseases in the future and offer new insight into the molecular basis of the initiation and progression of MM.

    Spectrophotometric Characterization of Plasticizer Migration in Poly(vinyl Chloride)-Based Artificial Leather

    Plasticized polyvinyl chloride (PVC) is one of the most commonly used material in the manufacture of artificial leather because of its flexibility, high durability and low cost. While hundreds of plasticizers have been produced to obtain PVC films and artificial leathers, it is important to choose the correct plasticizer to produce the desired products. Therefore, here five non-phthalate type plasticizers with three levels (40 phr, 60 phr and 80 phr); trioctyl trimelliate (TOTM), tributyl trimelliate (TBTM), dioctyl terephthalate (DOTP), dioctyl adipate (DOA) and dioctyl succinate (BIO) were used to prepare plasticized PVC films. They were studied in terms of their compabilities with PVC polymer chains, and their effects on mechanical properties of PVC films. Although, linear plasticizers e.g. BIO and DOA have higher compabilities with PVC, a branched plasticizer e.g. TOTM showed higher mechanical properties. Moreover, a new spectrophotometric method for determination of plasticizer migration from PVC polymer matrix to the surface of artifical leather was developed. According to lightness change on the surface of PVC artifical leather before and after heat treatment at 70 °C, the migration rates of used plasticizers increase in this order: TOTM < TBTM < DOTP < BIO < DOA. © 2020 Elsevier B.V.

    Epo Mediates Neurotrophic, Neuroprotective, Anti-Oxidant, and Anti-Apoptotic Effects Via Downregulation of Mir-451 and Mir-885 in Sh-Sy5y Neuron-Like Cells

    Erythropoietin (EPO) is a neuroprotective cytokine, which has been applied in several animal models presenting neurological disorders. One of the proposed modes of action resulting in neuroprotection is post-transcriptional gene expression regulation. This directly brings to mind microRNAs (miRNAs), which are small non-coding RNAs that regulate gene expression at the post-transcriptional level. It has not yet been evaluated whether miRNAs participate in the biological effects of EPO or whether it, inversely, modulates specific miRNAs in neuronal cells. In this study, we employed miRNA and mRNA arrays to identify how EPO exerts its biological function. Notably, miR-451 and miR-885-5p are downregulated in EPO-treated SH-SY5Y neuronal-like cells. Accordingly, target prediction and transcriptome analysis of cells treated with EPO revealed an alteration of the expression of genes involved in apoptosis, cell survival, proliferation, and migration. Low expression of miRNAs in SH-SY5Y was correlated with high expression of their target genes, vascular endothelial growth factor A, matrix metallo peptidase 9 (MMP9), cyclin-dependent kinase 2 (CDK2), erythropoietin receptor, Mini chromosome maintenance complex 5 (MCM5), B-cell lymphoma 2 (BCL2), and Galanin (GAL). Cell viability, apoptosis, proliferation, and migration assays were carried out for functional analysis after transfection with miRNA mimics, which inhibited some biological actions of EPO such as neuroprotection, anti-oxidation, anti-apoptosis, and migratory effects. In this study, we report for the first time that EPO downregulates the expression of miRNAs (miR-451 and miR-885-5p) in SH-SY5Y neuronal-like cells. The correlation between the over-expression of miRNAs and the decrease in EPO-mediated biological effects suggests that miR-451 and miR-885-5p may play a key role in the mediation of biological function.
