    Development of Si Doped Nano Hydroxyapatite Reinforced Bilayer Chitosan Nanocomposite Barrier Membranes for Guided Bone Regeneration

    Guided Bone Regeneration (GBR) is a widely used process for the treatment of periodontal defects to prevent the formation of surrounding soft tissue at the periodontal defect and to provide hard tissue regeneration. Recently GBR designs have focused on the development of resorbable natural polymer-based barrier membranes due to their biodegradability and excellent biocompatibility. The aim of this study is to fabricate a novel bilayer nanocomposite membrane with microporous sublayer composed of chitosan and Si doped nanohydroxyapatite particles (Si-nHap) and chitosan/PEO nanofiber upper layer. Bilayer membrane was designed to prevent epithelial and fibroblastic cell migration and growth impeding bone formation with its upper layer and to support osteogenic cell bioactivity at the defect site with its sublayer. Microporous and nanofiber layers were fabricated by using freeze-drying and electrospinning techniques respectively. The effect of Si-nHap content on the morphological, mechanical and physical properties of the composites were investigated using SEM, AFM, micro-Ct, compression test, water uptake capacity and enzymatic degradation study. Antimicrobial properties of nanocomposite membranes were investigated with tube dilution and disk diffusion methods. In vitro cytotoxicity of bilayer membranes was evaluated. Saos-2 and NIH/3T3 proliferation studies were carried out on each layer. In vitro bioactivity of Saos-2 and NIH/3T3 cells were evaluated with ALP activity and hydroxyproline content respectively. Results showed that Si-nHap incorporation enhanced the mechanical and physical properties as well as controlling biodegradability of the polymer matrix. Besides, Si-nHap loading induced the bioactivity of Saos-2 cells by enhancing cell attachment, spreading and biomineralization on the material surface. Thus, results supported that designed bilayer nanocomposite membranes can be used as a potential biomaterial for guided bone regeneration in periodontal applications.

    Tekstil Endüstrisinde Bütünleşik Membran Sistemi ile Su Geri Kazanımı ve Hibrit İleri Oksidasyon/membran Filtrasyonu ile Konsantrelerin Arıtımı ve Yönetimi

    Bu çalışmada, etkinliği bilinen UF/NFsıkı/TO membran kombinasyonu ile tekstil atıksularının arıtımının yanısıra %75-80 su geri kazanım oranı dahilinde oluşan membran konsantrelerinin, bütünleşik ileri oksidasyon prosesi (Fenton ve foto-Fenton (UVA-365–UVC-254))/batık UF sistemi ile daha önce belirlenen optimum işletme şartlarında oksidasyona tabi tutularak yönetimi gerçekleştirilmiştir. Yapılan deneysel çalışmalar sonucunda, UF/NFsıkı/TO membran konfigürasyonu ile arıtımı sonucunda oluşan konsantre akımlarının Fenton, foto-Fenton (UVA-365) ve (UVC-254) prosesleri/batık UF sistemi ile oksidasyonu sonrası, ardışık işletimli foto-Fenton (UVC-254)/UH050 hibrit prosesinin 9.88 H2O2/TOK ve 7.27 H 2 O 2 /Fe 2 + oranlarında, 44 L/m 2 .sa’lik süzüntü akısında %86 KOİ giderimi ile KOİ değeri 167 mg/L’ye ve %74.2 TOK giderimi ile TOK değeri 104 mg/L’ye kadar düşürülerek en iyi performans gösteren yenilikçi proses olduğu sonucuna varılmıştır. Oluşan membran konsantrelerin oksidasyonu sonrasında kalan konsantrenin tehlikelilik özellikleri, bertaraf/geri kazanım yöntemleri ve yeniden kullanım alternatifleri değerlendirildiğinde tekstil endüstrisi atıksularının foto-Fenton (UVA-365) ile arıtımı sonucunda oluşan konsantre haricinde diğer tüm konsantrelerin tehlikeli özelliğine sahip olduğu görülmüştür. Konsantrelerin bertaraf/geri kazanım yöntemleri ayrıntılı olarak değerlendirildiğinde, gerekli düzenlemeler (nem içeriği, susuzlaştırma, uygun debi ve/veya karışım oranları) yapıldığı takdirde bu konsantrelerin düzenli depolama, yakma ve merkezi atıksu arıtma sistemine verme gibi seçeneklerle bertaraf edilebileceği sonucuna varılmıştır.

    Xenograft Tumor Volume Measurement in Nude Mice: Estimation of 3d Ultrasound Volume Measurements Based on Manual Caliper Measurements

    Objectives: Volume measurement of subcutaneous xenograft tumors in nude mice models is an important metric to assess tumor growth or response to therapy. Manual calipers are widely used for this purpose. But the measurements with manual calipers may be inaccurate. Contrarily, three-dimensional (3D) ultrasonographic measurements give reliable and accurate tumor volume calculation. We aim to; evaluate the accuracy of common four formulas given in the literature to estimate xenograft tumor volumes based on manual caliper measurements and offer a new coefficient for a better estimation of the tumor volumes. Patients and Methods: Detailed manual diameter measurements of xenograft tumors were in 14 nude mice performed using Vernier caliper. Tumor volumes were calculated using the suggested formulas in the literature based on manual measurements. 3D ultrasound volume measurements were performed on same xenograft tumors using high resolution Vevo 2100 imaging system. To propose a new coefficient; means of ratio between manual and ultrasound volume measurement values were used. Also, data set was divided into two subgroups as tumor volume under 800 mm3 and over 800 mm3. New coefficients for each subgroup were defined. Results: Only with prolate ellipsoid formula there was no statistically significant difference between volume measurements with two methods (p=0,24). Our proposed formula (0,45 L*W*H) could estimate tumor volumes as good as prolate ellipsoid formula. Coefficient 0,35 and 0,81 in the same formula were found efficient in the selected subgroups. Conclusion: Using one common coefficient/formula for tumor volume estimation in any tumor size can be inaccurate. Appropriate coefficient should be chosen according to the dataset worked with.

    Effect of Some Physical, and Chemical Variables on Flocculation and Sediment Behaviour

    Effect of some chemical and physical variables on the settling rate, final sediment height, sediment viscosity and supernatant turbidity of a clay sample was studied using various polyacrylamide type flocculants. Increasing flocculant concentration significantly increased both the settling rate and sediment viscosity. More importantly, changes in the final sediment, height, hence the packing density, was minimal for all the conditions tested once the sediment was allowed to consolidate. Also, the mode of addition of the polymer, at once or continuous, did not seem to affect any of the parameters measured. Conditioning time seemed to alter the settling rate at low polymer concentrations, but had no effect at high polymer concentrations. However, increasing the conditioning time caused a decrease in the sediment viscosity. Different types of the polyacrylimides generated different settling rates at a given concentration, but the final sediment height was nearly independent of polymer type.

    Amphiphilic poly(methyl methacrylate)-b-poly(acrylic acid) (PMMA-b-PAA) copolymers with 4 and 6 arms were synthesized by Atomic Transfer Radical Polymerization (ATRP). The polymers were characterized by GPC, H-NMR and FTIR analysis. Polymer samples synthesized at various molecular weights between 18-80 kDa were investigated in terms of maximum loading capacity and critical micelle concentration by fluorescence spectroscopy. 4 arm PMMA-b-PAA (MW 30 kDa) was loaded with hydrophobic model drug indomethacin with an extremely high drug loading efficiency (22.5% drug content). Drug loaded polymer formed a stable and biodegradable complex in aqueous medium.
