    Kentlerin Depreme Dirençliliğinin Bina Bazlı Bölgesel Risk Dağılımı Yöntemi İle İncelenmesi: İzmit Kenti Örneği

    Türkiye, yıkıcı deprem üretme potansiyeli yüksek fay hatlarının yer aldığı bir bölgede konumlanmaktadır. Tarih boyunca, Anadolu yarımadasında meydana gelen depremler büyük can ve mal kayıplarına sebep olmuştur. Bu bağlamda, nüfusun ve sanayileşmenin çok yoğun olduğu Marmara Bölgesinde deprem risk değerlendirme çalışmalarının yoğunlaşması ve gerekli önlemlerin alınması büyük önem arz etmektedir. Bu çalışmada, 6306 sayılı \"Afet Riski Altındaki Alanların Dönüştürülmesi Hakkında Kanun\" kapsamında tanımlanan binaların bölgesel deprem riski dağılımının belirlenmesi için riskli yapıların tespitine ilişkin esaslar başlığı altında öngörülen basitleştirilmiş yöntemler kullanılarak Kocaeli ili İzmit ilçesinde bulunan 19940 bina incelenmiştir. Söz konusu yöntem, hızlı sokak taraması prensiplerini dikkate alarak bölgesel deprem risk önceliklendirmesini hedeflemektedir. Saha incelemelerinden elde edilen sonuçlar, nüfus yoğunluğu ve dağılımı, acil toplanma alanı dağılımı, toplam yapı alanı gibi parametrelerle beraber değerlendirilerek İzmit ilçesinde deprem risk öncelikli bölgelerin belirlenmesine çalışılmıştır.

    Günümüzde kütüphaneler, değişen teknoloji ve yeniliklerden etkilenen kurumlar arasında yer almaktadır. Yapay zeka teknolojilerinin popüler hale gelmesi, kütüphane hizmetlerini de dönüştürmeye başlamıştır. Bu araştırmada, Türkiye’deki üniversite kütüphanelerinin yapay zeka teknoloji ve uygulamalarının gelişim sürecinde yapmış olduğu ve yapmayı planladığı düzenlemeleri tespit etmek ve ilgili döneme özel geliştirdikleri hizmetleri belirlemek amacıyla bir anket uygulanmıştır. Anket, Türkiye’deki 208 üniversite kütüphanesinden 111 üniversite kütüphanesi yöneticisinin katılımıyla gerçekleştirilmiştir. Verilerin analizi ile üniversite kütüphanelerinin yapay zeka teknolojileri ve uygulamaları hakkındaki durumu, bilgi ve farkındalık düzeyleri belirlenmiş, eksik ve zayıf yönlerin geliştirilmesine yönelik önlemler ve öneriler sunulmuştur. İlgili araştırma, yapay zeka konusunda Türkiye’de üniversite kütüphanesi yöneticilerinden görüş ve öneri alarak gerçekleştirilen ilk ve en kapsamlı çalışmadır. Araştırma bulguları, üniversite kütüphanelerinin ChatGPT, Gemini, Grammarly vb. yapay zeka uygulamalarını belirli düzeyde kullandıklarını ancak yapay zeka ile ilgili kurumsal politika geliştirme, personele yetkinlik kazandırma ve planlama konularında ihtiyaçlarının olduğu ortaya çıkmıştır.

    Zinc Borate Chemical Garden and Zinc Borate Powders From Tincal Mineral and Zinc Sulfate Heptahydrate

    The formation of the first membrane, the swelling of the crystal by incoming water from the semipermeable membrane and the formation of irregular shaped branches were observed by optical microscopy, when zinc sulfate heptahydrate crystals were immersed in saturated borax solution. The powders obtained by mixing dilute aqueous borax and zinc sulfate solutions had B, O, Na, S and Zn elements. Presence of $Na Zn_{1/2}B_4O_7.xH_2O$ was indicated by EDX analysis. The molar ratio of $B_2O_3/ZnO$ in powders was around 2. FTIR analysis indicated the ratio of absorbance values of asymmetric stretching vibrations of B(3)-O at 1351 cm-1 to that of B(4)-O at 1026 cm-1 increased with their heating time at 90ºC during their preparation. X-ray diffraction patterns indicated the presence of $Zn(OH)_2and Zn_4(OH)_6(SO_4)·4H_2O$. The zinc borate compounds in the powders were not crystalline since no sharp peaks related to zinc borates were present in x-ray diffraction diagram. There were two mass loss steps in TG curves of the powders. The first step at 150-350ºC and the second step at 700-950ºC were due to elimination of water and due to decomposition of sulfate ions respectively. The submicron powders were a mixture of zinc borate, $Zn(OH)_2, Zn_4(OH)_6(SO_4)·4H_2O and Na Zn_{1/2}B_4O_7.xH_2O$ and they could be used as lubricant additive due to their small particle size of 600 nm.

    Expression Patterns of M6a Rna Methylation Regulators Under Apoptotic Conditions in Various Human Cancer Cell Lines

    Background/aim: Cancer is a complex disease that involves both genetic and epigenetic factors. While emerging evidence clearly suggests that changes in epitranscriptomics play a crucial role in cancer pathogenesis, a comprehensive understanding of the writers, erasers, and readers of epitranscriptomic processes, particularly under apoptotic conditions remains lacking. The aim of this study was to uncover the changes in the expression of m6A RNA modifiers under apoptotic conditions across various cancer cell lines. Materials and methods: Initially, we quantified the abundance of m6A RNA modifiers in cervical (HeLa and ME180), breast (MCF7 and MDA-MB-231), lung (A549 and H1299), and colon (Caco-2 and HCT116) cancer cell lines using qPCR. Subsequently, we induced apoptosis using cisplatin and tumor necrosis factor-alpha (TNF-α) to activate intrinsic and extrinsic pathways, respectively, and assessed apoptosis rates via flow cytometry. Further, we examined the transcript abundance of m6A RNA modifiers under apoptotic conditions in cervical, breast, and lung cancer cell lines using qPCR. Results: Overall, treatment with cisplatin increased the abundance of m 6A modifiers, whereas TNF-α treatment decreased their expression in cervical, breast, and lung cancer cell lines. Specifically, cisplatin-induced apoptosis, but not TNF-α-mediated apoptosis, resulted in decreased abundance of METTL14 and FTO transcripts. Additionally, cisplatin treatment drastically reduced the abundance of IGF2BP2 and IGF2BP3 readers. Conclusion: These results suggest that the differential response of cancer cells to apoptotic inducers may be partially attributed to the expression of m6A RNA modifiers.

    Accessibility in Intensive Care Units: a Qualitative Study on Exploring Architects’ Perspective

    Thisstudyaddresseshealthcaredesigners’perspectivesconcerningthearchitectural features within the Intensive Care Unit (ICU) environments that can impact visual and physical access to patients. In line with patient-centered approaches, providing accessible environments in ICUs is becoming increasingly critical for healthcare providers. The existing literature suggests various architectural features to influence levels of access to patients. How architects prioritize these features and translate them into the configuration of ICU environments has not been explored extensively. A series of semi-structured interviews were conducted to understand the perspectives of healthcare architects in the context of Turkey. The interviews were conducted with twelve participants with recent experiences in ICU design. The research followed a thematic analysis to link the qualitative data with the participants’ drawings that emerged during interviews. Five essential themes emerged, including: “Unit Model,” “Unit Layout,” “Unit Size,” “Bed Position,” and “Transparent Material.” The participants implied configurational models, including “open ward” and “single-patient room,” to facilitate high levels of accessibility. Beyond the key decisions concerning layouts, the participants also emphasized the strategic use of transparent materials, which was considered critical in establishing visual access within units. The findings suggest that healthcare architects mostly favor open wards as a suitable model to provide high levels of physical access by decreasing nurses’ walking distances during shifts and visual accessibility by enhancing nurses’ capacity to supervise the patients within ICU environments. The findings can advance our understanding of how the issue of access is formulated and implemented in ICU settings.

    İzmir'in bir dağ köyü: Yamanlar ve Yamanlar Yörükleri üzerine etnotarihsel bir inceleme

    This study examines the history, language, social, and economic transformation of the Yuruks living in the Yamanlar Village located in the Karsiyaka district, where there is relatively little information in the literature despite its proximity to the Izmir city center. Information collected from the Ottoman Era cemetery adjoining the village reveals that the Yamanlar Yuruks have lived here for at least 400 years. Mount Yamanlar, where the village is located, is of great importance because of its ancient history, as it witnessed the founding of Izmir (Smyrna). The social and economic lifestyle in the village has transformed as the city expanded toward residential areas atop Mount Yamanlar. The loss of traditional livelihood practices challenged the villagers and severely influenced their customs, which might be considered valuable pieces of cultural heritage. In this study, we conducted fieldwork, during which various data collection methods and techniques were employed to describe daily life in the village. Within the scope of this study, semi-structured and unstructured interviews were conducted with the local people, and an oral historical study was carried out. With this method, we aimed to present an ethnohistorical description of the Yamanlar Village by trying to establish a balance between the analysis of both written documents and oral data to make sense of the social and economic change it has undergone and to contribute to the literature on Izmir Yuruks.

    Contradicting Parochial Realms in Neighborhood Parks: How the Park Attributes Shape Women’s Park Use

    Neighborhood parks are significant green public spaces located in close social and geographical proximity to homes to maintain individual and public health. However, some people do not use the nearest parks, but those with other socio- spatial attributes that make them feel more “familiar”. This study argues that with their facilities, amenities and design, and the surrounding land uses, neighborhood parks do not only accommodate, but also define, regulate, and originate social relations among users. Thus, the design and planning of urban public spaces play a role in the emergence and maintenance of supportive and conflictual relations that lead to familiarity. The study answers two research questions: How do the park attributes shape and mediate the interpersonal relations among the park users? How do gender differences influence the parochial realms in parks? Data was collected through field observations and in-depth interviews with 33 female users of two neighborhood parks in a populous district of Izmir (Turkey). Results state that women’s park visits were related to their gendered roles and responsibilities. Yet their responses point to challenges emerging from physical and social attributes of parks and park surroundings which lead to negotiations to protect their individual or group’s privacy (parochial realm) in neighborhood parks. Mainly, perceived threats to women’s parochial realm are men unaccompanied by child(ren), and exposure to the male gaze. The study highlights the importance of investigating these attributes of neighborhood parks for developing research and public policies to improve women’s presence and perceived safety in public settings.

    Fabrication of Superhydrophilic Teos-Lactic Acid Composite Films and Investigation of Biofouling Behaviour

    Phytoplankton and diatom microalgae species cause biofouling by adhering to the surfaces, especially in closed cultivation systems such as tubular photobioreactors. This biofilm formation blocks the sunlight; after harvesting, it is necessary to clean the reactor. This cleaning process causes loss not only for time and finance but also in terms of environmental pollution due to using toxic chemicals and excess water usage. This study aimed to investigate the reduction of the microorganism cell adhesion on the hybrid surface. To succeed in this, the composite surface of tetraethoxysilane (TEOS) and lactic acid (LA) was prepared by the sol-gel process. Then the hybrid surfaces were coated on glass slides by the dip coating method. The wettability performance of the TEOS-LA hybrid surface was investigated using contact angle measurement and light transmittance. The wettability result showed that the superhydrophilic surface having 54 mJ/m2 of surface free energy values was obtained. An increase in the lactic acid content of the composite films increased the surface free energy (SFE) values decreasing the water contact angle. A pencil hardness test characterized the mechanical strength of the surfaces, and it was determined that the hardness of the composite films was decreased by increasing the LA content of the composite films. Resultantly, it is found that the TEOS-LA superhydrophilic composite film reduces the adhesion of microalgae.

    Epithelial-Mesenchymal Transition as a Potential Route for Dapt Resistance in Breast Cancer Cells

    Objectives: Notch is a conserved pathway involved in cell- fate determination and homeostasis. Its dysregulation plays a role in poor prognosis and drug resistance in breast cancer. Targeting Notch signaling via inhibition of the gamma- secretase complex is in the spotlight of modern cancer treat- ments. Gamma-secretase inhibitors (GSI) have shown suc- cessful clinical activity in treating cancers, yet the possible resistance mechanism remains unstudied. Modeling the resistance and understanding culprit molecular mechanisms can improve GSI therapies. Accordingly, the aim of this study is to generate and analyze GSI-resistant breast cancer cells. Methods: Gradually increasing doses of DAPT, a well-known GSI, were applied to MCF-7 breast cancer cell lines to generate resistance. Cell viability, migration and gene expressions were assessed by MTT, wound healing and qRT-PCR analyses. Results: DAPT-resistant MCF-7 cells exhibited abnormal expression of Notch receptors, Notch targets (HES1, HES5, HEY1), and epithelial-mesenchymal transition (EMT) markers (E-cadherin, ZO-1, SNAIL2, N-cadherin) to overcome the continuous increase in DAPT toxicity by increased migration through mesenchymal transition. Conclusions: This study prospects into the role of EMT in the potential resistance mechanism against DAPT treatment for breast cancer cells. Complementary targeting of EMT should be investigated further for a possible effect to potentiate DAPT’s anti-cancer effects.

    Improvement of Strength Characteristics of a Highly Plastic Expansive Soil by Fly Ash

    Highly plastic expansive clays swell or shrink due to change in moisture content and they often have very low bearing capacity. Construction of engineering structures particularly pavements and lightweight buildings on problematic soils such as highly plastic expansive clays may create severe structural problems due to poor engineering properties of that kind of soil. This study focused on the influence of fly ash on strength properties of highly expansive clays which are problematic soils due to their undesirable engineering characteristics such as high plasticity index, liquid limit, swelling and shrinkage characteristics, and low bearing capacity. Atterberg’s limit tests and hydrometer tests were conducted to obtain consistency limits and grain-size distribution of the highly plastic expansive clay. Standard proctor tests for clay samples with different fly ash contents such as 10%, 15%, and 20% were performed to determine maximum dry densities and optimum water contents of the mixtures of clay and fly ash. The clay and fly ash mixtures were prepared at optimum water content and maximum dry density for unconfined compression strength (UCS) and California Bearing Ratio (CBR) tests. Effects of fly0ash content on the strength characteristics of a highly plastic expansive clay were investigated through unconfined compression and CBR tests. The experiment results showed that fly0ash is a promising additive to enhance strength characteristics of highly plastic expansive clays. An approximately 66% increase in UCS and CBR values was observed with the addition of 20% fly ash into the clay mixture in weight. © 2022, Sakarya University. All rights reserved.
