    Determination of Resistance Levels To Clavibacter Michiganensis Subsp. Michiganensis in Some Solanum Species

    Clavibacter michiganensis subsp. michiganensis (Cmm), is a devastating bacterial disease agent causing bacterial wilt and canker in tomatoes. There is no definitive solution to prevent yield losses by Cmm in tomatoes. Moreover, there is currently no commercially successful Cmm resistant tomato cultivar on the market. Therefore, we aimed to determine the tolerance level of some tomato accessions to Cmm in the present study. For this purpose, we screened seven tomato accessions representing four species (Solanum arcanum, S. habrochaites, S. pennellii, and S. peruvianum) from Peru, Ecuador, and Mexico against the highly virulent isolates Cmm-244 and Cmm-9. A root immersion method was used to identify new sources of resistance to this important disease. Two accessions, S. habrochaites LA1777, and S. arcanum LA2157 were found to be moderate and highly tolerant, respectively, and could serve as tolerance resources for tomato breeding in Türkiye. These materials can also be investigated more extensively to determine their intrinsic Cmm tolerance mechanism.

    Thrips parazitoiti Ceranisus (Hymenoptera: Eulophidae) cinsinin morfolojik yapı ve 28S D2 rDNA temelinde Türkiye’deki güncel taksonomik durumları

    Çalışma ile Mayıs 2006 ile Mayıs 2007 tarihleri arasında Güneydoğu Anadolu Bölgesi ve Akdeniz Bölgesi’nin doğusunda bulunan Ceranisus (Hymenoptera: Eulophidae) cinsi ve benzer diğer cinslerin genetik analizleri yapılarak, bu verilerin diğer thrips parazitoitlerinin morfolojik yapılarıyla mukayese edilip, türlerin kesin teşhislerinin yapılması amaçlanmıştır. Bu kapsamda, Hatay, Gaziantep, Adıyaman ve Şanlıurfa’da 38 farklı noktada örnekleme yapılmış ve 20 örnekleme noktasında toplam 143 adet (30 erkek; 113 dişi) Ceranisus cinsine giren tür toplanmıştır. Populasyon morfolojik özelliklerine göre 13 farklı gruba ayrılmıştır. Bu türlerin filogenetik ilişkileri DNA dizileri kullanılarak verilmiştir. Toplam 15 tür 28S D2 ribosomal DNA bölgesi kullanılarak analiz edilmiştir. Analiz sonucunda morfolojik olarak ayrılan 13 farklı tür grubundan 3 tanesinin genetik olarak farklı olduğu görülmüştür. Diğer türlerden ise 4 adetinin C. nr. lepidotus grubu oluşturdukları, 5 adet C. nr. bozovaensis’in ise kendi aralarında 2 farklı grup oluşturdukları belirlenmiştir. Sonuç olarak 3 adet yeni Ceranisus türü, bunun haricinde 3 adet farklı türü oluşturabilecek popülasyon genetik olarak belirlenmiştir.

    Application of Est-Ssrs To Examine Genetic Diversity in Eggplant and Its Close Relatives

    Within the genus Solanum, the term 'eggplant' encompasses several cultivated species that are used for food and, to a lesser extent, for medicine. Th e use of one common name to describe more than one species and the existence of many related wild species have led to taxonomic confusion which, in turn, have complicated analyses of evolutionary relationships and genetic diversity within this groups of species. A further challenge for eggplant research is that, despite the fact that the use of molecular markers for phylogenetic studies is well-established, very few studies have described the development of new markers for eggplant. In our work, genic microsatellite (SSR) markers were identified from an expressed sequence tag library of S. melongena and used for analysis of 47 accessions of eggplant and closely related species. Th e markers had very good polymorphism in the 18 species tested including 8 S. melongena accessions. Moreover, genetic analysis performed with these markers showed concordance with previous research and knowledge of eggplant domestication. Th ese markers are expected to be a valuable resource for studies of genetic relationships, fingerprinting, and gene mapping in eggplant.

    Quantitative Trait Loci (qtl) Analysis for Antioxidant and Agronomically Important Traits in Tomato (lycopersicon Esculentum)

    Tomato is one of the most widely produced and consumed vegetable crops worldwide. Plant breeders have usually focused on improvement of horticulturally important traits such as yield, fruit size, shape and colour. With increased attention on human health, however, plant breeders also consider the improvement of health-related traits of fruits and vegetables such as antioxidant characters. In the present study, genes controlling both health-related and horticulturally important traits were mapped in the tomato genome using 152 Lycopersicon hirsutum BC2F2 individuals. For this aim, all plants were phenotypically and genotypically characterised and a total of 75 QTLs were identified for all traits. Of the 75 QTLs, 28 were identified for 5 antioxidant traits including total water soluble antioxidant capacity, vitamin C, total phenolics, total flavonoids, and lycopene contents, and 47 QTLs were identified for 8 agronomic traits including fruit weight, external and internal fruit colour, fruit firmness, fruit shape, stem scar size, locule number, and wall thickness. Markers linked with these QTLs can be used in marker assisted selection (MAS) for improvement of elite tomato lines. © TÜBİTAK.

    Preliminary Phylogeny of the Thrips Parasitoids of Turkey Based on Some Morphological Scales and 28s D2 Rdna, With Description of a New Species

    Species of the Ceranisus thrips-attacking genus are difficult to distinguish morphologically. The phylogenetic relationships within the Ceranisus species were explored using nucleotide sequences of the 28S D2 expansion region of the rDNA gene. Bayesian, maximum likelihood, and parsimony inference methods were employed to construct the phylogenetic relationships. Principal component analysis on the Turkish species of Ceranisus, namely antalyacus, menes, bozovaensis, hirsutus, planitianus (a new record for Turkey), pacuvius, and a new species, provided supporting evidence. All known data concerning hosts and geographical distribution are presented. A new species, C. onuri O. Doganlar, sp.n., was described from Turkey. © TÜBİTAK.

    Identification of Potato Y Potyvirus (pvy°) Resistance in Wild and Cultivated Tomatoes

    Potato Y potyvirus (PVY) is an important plant pathogen worldwide that infects and causes yield losses in the family Solanaceae including potato (Solarium tuberosum), pepper (Capsicum spp.), tomato (S. lycopersicum), and tobacco (Nicotiana tabacum). In this study, 20 different tomato accessions representing 6 different species were mechanically inoculated with PVY°. The plants were scored visually for symptoms and then tested for presence of the virus 2-4 weeks after inoculation by ELISA. The results were variable. Most wild species of tomato sustained PVY° replication in inoculated leaves. Some of the wild species showed an immune response, while some became systemically infected. Inoculation and analysis of F2 populations suggested that the resistance is controlled by a single recessive gene in different wild species.
