    Development of Cellulose Acetate Based Antimicrobial Food Packaging Materials for Controlled Release of Lysozyme

    Antimicrobial packaging materials were obtained by incorporation of lysozyme into cellulose acetate (CA) films. In order to achieve controlled release of lysozyme, the structure of the films was changed from highly asymmetric and porous to dense by modulating the composition of the initial casting solution. The highest release rate, soluble lysozyme activity and antimicrobial activity were obtained with the film prepared from 5% CA solution including 1.5% lysozyme. Increasing CA content in the casting solution decreased the porosity of the films, hence, reduced the release rate, maximum released lysozyme activities and the antimicrobial activities of the films. In contrast, immobilized lysozyme activities and the tensile strength of the films increased. The incorporation of lysozyme did not cause significant reductions in tensile strength and elongation at break values except in films prepared with 15% CA. This study showed the good potential of asymmetric CA films to achieve controlled release in antimicrobial packaging.

    Water Vapour Barrier Performance of Corn-Zein Coated Polypropylene (pp) Packaging Films

    The novel film structure of corn-zein coated on polypropylene (PP) synthetic film for packaging industry was developed to examine the feasibility of resulting coated films as an alternative water barrier performance for food packaging. The effects of coating formulation (solvent, corn-zein, plasticizer concentration and plasticizer type) on final properties of films were observed. Corn-zein is the most important protein of corn and has good film forming property. Composites structures of PP films coated with corn-zein were obtained through a simple solvent casting method. Polyethylene glycol (PEG) and glycerol (GLY) were used as plasticizer to increase film flexibility. Statistical analysis based on full factorial design was performed to observe coating formulation effects. The high water vapour barriers were obtained for films coated with coating formulation consisting of higher amounts of corn-zein plasticized by GLY. The lower glass transition temperatures (T g) of films were obtained by plasticization of films and T g decreased by increasing plasticizer content. The statistical analysis defined the key parameters of coating formulation that had major effects on the final properties of coated PP films as corn-zein, plasticizer concentration and plasticizer type. In conclusion, corn-zein coatings could have potential as an alternative to conventional synthetic polymers used in composite multilayer structures for food packaging applications.

    Separation of Whey Components by Using Ceramic Composite Membranes

    Ceramic supports were prepared from alumina powder and dip-coated with zirconia sol. The ceramic composite membranes prepared were characterized with respect to their microstructure/pore structures. The supports were 40% porous of which 87% were open pores. The average particle size of the sol particles was 35 nm. The prepared membrane has good protein lactose separation properties with a relatively high protein content (PR ∼80%) and with relatively low lactose retention (LR ∼7%). The permeate flux value was around 40 l/m2h. These results indicate the possibility of the preparation of ceramic composite membranes for separation of whey components with higher yields.

    Optimisation of the Effect of Colemanite as a New Synergistic Agent in an Intumescent System

    An intumescent system including ammonium polyphosphate (APP) as an acid source and blowing agent, pentaerythritol (PER) as a carbonific agent and colemanite as a synergistic agent is used to enhance flame retardancy of polypropylene (FR-PP). In order to investigate the synergism between colemanite and the flame retardant materials (APP and PER), D-optimal mixture design was employed. The limiting oxygen index (LOI) and amount of residue (AoR) were accepted as response 1 and response 2, respectively. Applying D-optimal strategy, 18 experiments were performed. Filler content was fixed at 30 wt% of total amounts of flame retardant PP composites. Constraints were determined according to the ratio of APP/PER ranging between 1 and 3. Statistical analysis of the cubic model revealed that lack of fit (LoF) was not significant for the cubic and linear model for both responses. The model suggested an optimum composite formulation with concentration levels 65% of APP, 28% of PER and 7% of colemanite that gives an LOI of 40.3. The experimental LOI and AoR of optimum formulation were achieved as 39.3 and 21.4 with 2.5% and 2.2% errors, respectively.

    Partial Purification of Hen Egg White Lysozyme by Ethanol Precipitation Method and Determination of the Thermal Stability of Its Lyophilized Form

    Lysozyme was partially purified from hen egg white by precipitation of non-lysozyme protein impurities during incubation in the prence of ethanol. The thermal stability of the obtained partially purified enzyme was also characterized. The incubation of diluted egg white for 2-8 h in the presence of 20% ethanol was not very effective for the partial purification of lysozyme by precipitation of major egg white proteins; however, 4- to 6-h or 6-h to 8-h incubation of diluted egg white in the presence of 30% and 40% ethanol could be employed more effectively for partial purification of lysozyme. Without applying the incubation period, the highest specific activity was obtained by the treatment of egg white with 40% ethanol. Thus, ethanol at this concentration could be used for a continuous process of partial purification. For batch lysozyme purification, on the other hand, incubation in the presence of 30% ethanol was more appropriate. The activities and protein contents of dialyzed and lyophilized enzymes obtained by 6 h-incubation in the presence of 20%, 30%. and 40% ethanol precipitations were 1878, 6669, and 6115 U/mg powder, and 0.98, 0.90, and 0.93 mg protein per mg powder, respectively. The ranges of thermal inactivation parameters, such as D (D80°C = 29.2-59 min, D90°c = 8.8-21 min) and z (Z80-90°c = 17.4-22.3 °C) values of the enzyme, clearly indicated the moderate and variable heat stability of lyophilized lysozymes obtained from different batches of egg white.

    Mechanical Properties of Hydroxyapatite Composites Reinforced With Hydroxyapatite Whiskers

    Sintering and mechanical behavior of pure and hydroxyapatite (HA) whisker reinforced HA composites were investigated in this work. Pure and composite samples were prepared by using a commercial powder and whiskers prepared by molten salt synthesis. The dry-pressed samples were sintered in the 800 and 1300°C range. The effect of whisker-addition on the mechanical properties of HA was investigated through compression and hardness testing. Compressive strength and fracture strain were observed to increase by the addition of whiskers.

    Preparation and Characterization of Polylactide-Hydroxyapatite Biocomposites

    In the present study, the preparation and characterization of polylactide-Hydroxyapatite(HA) composite films for biomedical applications have been studied. The effects of number of parameters such as polymer type, HA loading, surface modification and its concentration on the mechanical and microstructural properties of the composites were investigated. Poly-L-Lactide and 96/4 Poly(L-Lactide co D-Lactide) copolymer-HA composites containing 10-40 wt% HA particles have been prepared by solvent casting technique. The HA powder was synthesized by precipitation technique. Interfacial interactions between HA and polylactide polymer were modified to improve filler compatibility and mechanical properties of the composites by surface treatment of the HA with two different silane coupling agents; 3-aminopropyltriemoxysilane (AMPTES) and 3-mercaptopropyltrimethoxysilane (MPTMS) at three different concentrations(0.5-2 wt%). Silane treatment indicated improvements in the mechanical properties of the composites compared to the untreated HA loaded polylactide composites. Tensile test results showed that the maximum improvement in the mechanical properties of the composites was obtained for PLA composites containing 1 wt% aminofunctional silane treated HA and 0.5-wt% mercaptopropyltrimethoxy silane treated HA for PDLA composites. Scanning electron microscopy studies also revealed better dispersion of silane treated HA particles in the polymer matrix.

    Preparation of Ceramic Composite Membranes for Protein Separation

    Ceramic supports were prepared from fine alumina and zirconia powders by dry-pressing and slip-casting. These supports were heat treated in the 1100° - 1200°C temperature range and dip-coated with ceramic sols prepared from alkoxides by using sol-gel methods. The average sol particle sizes were measured as 3-7 nm for zirconia and 30-40 nm for alumina by laser scattering technique. The optimum heat treatment temperature range was determined as 500°-600°C for dip-coated membranes by using TGA (thermo gravimetric analysis) results. The microstructure of the ceramic composite membranes was investigated by SEM (Scanning electron microscope). The clean water permeability (CWP) of the membranes was tested by using deionized water in a filtration set-up. Separation experiments were performed with bovine serum albumin (BSA, Stokes diameter: 7 nm) solution and whey to determine the protein separation capacity of the composite membranes. The CWP of the dry pressed alumina supports heat treated at 1100°C was found to be higher than the permeability of the slip-casted zirconia supports heat treated at 1200°C. The protein retention of the slip-casted zirconia support was determined as 60% by using UV-Vis spectrophotometer (Abs. at 280nm). A protein retention value of 96% was achieved for whey after the modification of the support with dip-coating. The permeate flux was 40 L/m2hour for dry-pressed alumina support dip-coated with zirconia sol calcined at 500°C.
