Saccharomyces cerevisiae (budding yeast) has been extensively utilized to elucidate the molecular mechanisms underlying arsenic toxicity and resistance in eukaryotes. In this study, we applied a genomic DNA overexpression strategy to identify yeast genes that provide arsenic resistance in wild-type and arsenic-sensitive S. cerevisiae cells. In addition to known arsenic-related genes, our genetic screen revealed novel genes, including PHO86, VBA3, UGP1, and TUL1, whose overexpression conferred resistance. To gain insights into possible resistance mechanisms, we addressed the contribution of these genes to cell growth, intracellular arsenic, and protein aggregation during arsenate exposure. Overexpression of PHO86 resulted in higher cellular arsenic levels but no additional effect on protein aggregation, indicating that these cells efficiently protect their intracellular environment. VBA3 overexpression caused resistance despite higher intracellular arsenic and protein aggregation levels. Overexpression of UGP1 led to lower intracellular arsenic and protein aggregation levels while TUL1 overexpression had no impact on intracellular arsenic or protein aggregation levels. Simav Plain (Kutahya, Turkey) is one such area that was shown to have natural arsenic contamination in its waters and soils. Arsenic exposure through ingestion of edible crops cultivated in Simav Plain and associated health risks were assessed in this study. To achieve this objective, arsenic levels in 18 crop species were estimated based on measured soil arsenic concentrations. Individual and aggregate non-carcinogenic and carcinogenic risks associated with ingestion of arsenic-contaminated crops were then assessed with scenario-based deterministic point estimates and stochastic population estimates. Monte Carlo simulation was used for the estimation of population health risks. Accordingly, wheat was found as the highest-ranked crop specie for the both types of health risks, followed by tomato and potato, which are three of the most consumed crops in the region. The risk levels estimated in this study were relatively high, indicating consumption of crops grown in the plain may be posing significant health risks even at lower-bound estimates. Consuming wheat, tomato, potato, and their products from uncontaminated sources was found to reduce the aggregate risks up to 88% implicating the importance of proposing suitable management measures for similar risk-prone areas.Article Citation - WoS: 39Citation - Scopus: 47Quality of Groundwater Resources in Afghanistan(Springer Verlag, 2017) Hayat, Ehsanullah; Baba, AlperWater is the main source of energy production and economy in Afghanistan where agriculture accounts for more than 50% of the country’s gross domestic product (GDP). Access to safe drinking water is still a problem in the country, which has caused different health issues and even child mortality especially in rural areas. Groundwater is the main source of drinking water in the country. However, little knowledge is available about the quality of groundwater throughout the entire country, and its quality has not been investigated extensively yet like in other countries in the world. While most people think that consuming groundwater is a reliable and safe source of drinking water for health, the United Nations (UN) agencies report various kinds of waterborne diseases and even child mortalities due to drinking water quality in the country. In this article, significant geogenic and anthropogenic factors that play a vital role in groundwater contamination of the country are identified and explained. Different geogenic contaminations such as arsenic, fluoride, sulfate, and boron occur in several areas of Afghanistan that have a direct effect on human health. The water quality mapping for Afghanistan is completed for half of the country, which shows that groundwater is plagued by high levels of fluoride and arsenic in some areas. The water quality mapping of the other half of the country cannot be completed due to security concerns currently. Also, there are different kinds of waterborne diseases such as diarrhea, cholera, and dysentery that can be seen in different parts of the country because of anthropogenic activities which continuously deteriorate groundwater.Article Citation - WoS: 27Citation - Scopus: 31Statistical Analysis of Causes of Death (2005-2010) in Villages of Simav Plain, Turkey, With High Arsenic Levels in Drinking Water Supplie(Taylor and Francis Ltd., 2015) Gündüz, Orhan; Bakar, Coşkun; Şimşek, Celalettin; Baba, Alper; Elçi, Alper; Gürleyük, Hakan; Mutlu, Merdiye; Çakır, AyşeThe purpose of this research was to compare the causes of death in 5 villages situated in Simav Plain, Turkey, during 2005-2010 where different arsenic levels were detected in drinking water supplies. Since groundwater in Simav Plain had arsenic concentrations that ranged between 7.1 and 833.9 ppb, a two-phase research was formulated. In the first phase, public health surveys were conducted with 1,003 villagers to determine the distribution of diseases. In the second phase, verbal autopsy surveys and official death records were used to investigate the causes of death. In total, 402 death cases were found in the study area where cardiovascular system diseases (44%) and cancers (15.2%) were major causes. Cancers of lung (44.3%), prostate (9.8%), colon (9.8%), and stomach (8.2%) were comparably higher in villages with high arsenic levels in drinking water supplies. Concentrations higher than 0.01mg/L are detrimental to human health. We examined potential consequences for As contamination of freshwater resources based on hydrogeochemical investigations of geothermal waters in deep wells and hot springs collected from western Anatolia, Turkey. We analyzed samples for major ions and trace element concentrations. Temperature of geothermal waters in deep wells showed extreme ranges (40 and 230°C), while, temperature of hot spring fluids was up to 90°C. The Piper plot illustrated two dominant water types: Na-HCO3 - type for geothermal waters in deep wells and Ca-HCO3 - type for hot spring fluids. Arsenic concentration ranged from 0.03 to 1.5mg/L. Dominance of reduced As species, i.e., As(III), was observed in our samples. The Eh value ranged between -250 and 119mV, which suggests diverse geochemical conditions. Some of the measured trace elements were found above the World Health Organization guidelines and Turkish national safe drinking water limits. The variation in pH (range: 6.4-9.3) and As in geothermal waters suggest mixing with groundwater. Speciation analysis was conducted using an HPLC-ICP-MS system. Ingestion exposure to arsenic and associated health risks were assessed by combining the concentration and questionnaire data both for individual participants and the subject population. Inorganic arsenic dominated both in rice and bulgur but concentrations were about an order of magnitude higher in rice (160. ±. 38. ng/g) than in bulgur. Because participants also consumed more rice than bulgur, exposures were significantly higher for rice resulting in carcinogenic risks above acceptable level for 53% and 93% of the participants when the in-effect and the proposed potencies were used, respectively, compared to 0% and 5% for bulgur. An inorganic arsenic standard for rice would be useful to lower the risks while public awareness about the relation between excessive rice consumption and health risks is built, and bulgur consumption is promoted.Article Citation - WoS: 303Citation - Scopus: 336A Health Risk Assessment for Exposure To Trace Metals Via Drinking Water Ingestion Pathway(Elsevier Ltd., 2009) Kavcar, Pınar; Sofuoğlu, Aysun; Sofuoğlu, Sait CemilA health risk assessment was conducted for exposure to trace metals via drinking water ingestion pathway for Province of İzmir, Turkey. Concentrations of 11 trace metals were measured in drinking waters collected from 100 population weighted random sampling units (houses). The samples were analyzed in atomic absorption spectrometry for arsenic, and inductively coupled plasma-optical emission spectrometry for Be, Cd, Co, Cr, Cu, Mn, Ni, Pb, V, and Zn. Questionnaires were administered to a participant from each sampling unit to determine drinking water consumption related information and demographics. Exposure and risks were estimated for each individual by direct calculation, and for İzmir population by Monte Carlo simulation. Six trace metals (As, Cr, Cu, Mn, Ni, and Zn) were detected in >50% of the samples. Concentrations of As and Ni exceeded the corresponding standards in 20% and 58% of the samples, respectively. As a result, arsenic noncarcinogenic risks were higher than the level of concern for 19% of the population, whereas carcinogenic risks were >10-4 for 46%, and >10-6 for 90% of the population.Article Citation - WoS: 104Citation - Scopus: 115An Exposure and Risk Assessment for Fluoride and Trace Metals in Black Tea(Elsevier Ltd., 2008) Sofuoğlu, Sait Cemil; Kavcar, PınarExposure and associated health risks for fluoride and trace metals in black tea were estimated. Fifty participants were randomly recruited to supply samples from the tea that they drink, and self-administer a questionnaire that inquired about personal characteristics and daily tea intake. Analyzed trace metals included aluminum, arsenic, barium, cadmium, cobalt, chromium, copper, manganese, nickel, strontium, and zinc. Fluoride and four metals (Al, Cr, Mn, Ni) were detected in all samples while barium was detected only in one sample. The remaining metals were detected in >60% of the samples. Fluoride and aluminum levels in instant tea bag samples were greater than in loose tea samples (p < 0.05) while the differences in elemental concentrations of loose and pot bag tea samples were not significant. Median and 90th percentile daily tea intake rates were estimated as 0.35 and 1.1 l/day, respectively. Neither fluoride nor aluminum levels in black tea were found to associate with considerable risks of fluorosis and Alzheimer's disease, respectively. However, carcinogenic risk levels for arsenic were high; R > 1.0 × 10-6 even at the median level. According to sensitivity analysis, daily tea intake was the most influencing variable to the risk except for arsenic for which the concentration distribution was of more importance.
