Conference Object Citation - Scopus: 2Preparation and Characterization of Hydroxyapatite/Calcium Phosphate Powders(Trans Tech Publications, 2004) Şimşek, Deniz; Çiftçioğlu, Rukiye; Harsa, ŞebnemThe preparation of calcium phosphate based powders through chemical precipitation in order to determine the optimum conditions for the preparation of stoichiometric, thermally stable hydroxyapatite (HA) powders was investigated in this work. The effects of parameters like pH, aging time, and heat treatment temperature on the phase structure and morphology of the powders were investigated by SEM and XRD.HA was prepared at a Ca/P ratio of 1.67. The precipitation pH had a profound influence on the morphology and crystallite size of the powders in the 4-11 pH range. Relatively large 5 mum prismatic particles were obtained at low pH whereas the powders were formed from agglomerates of nano sized particles at high pH. Powders had a HA dominant phase structure above a pH of 8. Phase pure HA powders were obtained with close to theoretical yields at pH = 10 and 11. The crystallite sizes of the powders varied from about 30 nm at 90degreesC to 50-90 mn at 1250degreesC with the smallest crystallite sizes obtained for pH 10 and 11 powders at 1250degreesC. The crystallite size increased significantly with aging temperature.Article Citation - WoS: 24Citation - Scopus: 35Physicochemical and Rheological Properties of Rice-Based Gluten-Free Blends Containing Differently Treated Chickpea Flours(Academic Press Inc., 2018) Kahraman, Gökçen; Harsa, Şebnem; Lucisano, Mara; Cappa, CarolaThis study focused on the evaluation of the physicochemical and rheological properties of chickpea flours and blends obtained by partially substituting rice flour (25 g/100 g) with raw, roasted and dehulled chickpea flour. The characteristics of the resultant doughs were evaluated. In comparison with rice flour, blends containing chickpea flours exhibited high protein and fat content, a reduced retrogradation tendency (setback values of 404–415 vs. 479 Brabender Unit) and a higher foaming capacity and stability, which can be beneficial for their use in baked food formulations. However, roasting decreased foaming capacity and stability. Even if the rheofermentographic test evidenced a slight reduction in dough development, high CO2 retention capacity (≥ 98%) and similar-to-lower leavening times were observed for doughs containing chickpea flours. Incorporating chickpea flours also caused an increase in the viscous and elastic moduli of rice-based doughs, resulting in a good structuring of the dough. Microcapsules were analyzed for physicochemical characteristics including morphology, particle size, moisture content, water activity, dissolution time, and color properties. Results revealed that microcapsules were spherical in shape and obtained particle sizes between 5 and 160 µm, with an average size of around 50 µm. Blending pullulan with WP provided enhanced survival of probiotic bacteria during spray drying with a final viable cell number of 8.81 log CFU/g of microcapsule. Encapsulated probiotics were also found to have significantly (p ≤ 0.05) higher survived cell numbers compared to free probiotics under detrimental gastrointestinal conditions. Moreover, dissolution analysis suggested that protein-polysaccharide powdered microcapsules showed pH-sensitive dissolution properties in simulated gastric juice and simulated intestinal juice.Article Citation - WoS: 24Citation - Scopus: 25Investigation of the Structure of Alpha-Lactalbumin Protein Nanotubes Using Optical Spectroscopy(Cambridge University Press, 2014) Tarhan, Özgür; Tarhan, Enver; Harsa, ŞebnemAlpha-lactalbumin (α-la) is one of the major proteins in whey. When partially hydrolysed with Bacillus licheniformis protease, it produces nanotubular structures in the presence of calcium ions by a self-assembly process. This study presents investigation of α-la protein structure during hydrolysis and nanotube formation using optical spectroscopy. Before spectroscopic measurements, nanotubes were examined with microscopy. The observed α-la nanotubes (α-LaNTs) were in the form of regular hollo strands with a diameter of about 20 nm and the average length of 1 μm. Amide and backbone vibration bands of the Raman spectra displayed remarkable conformational changes in α and β domains in the protein structure during nanotube growth. This was confirmed by the Fourier-transform infrared (FTIR) spectroscopy data. Also, FTIR analysis revealed certain bands at calcium (Ca++) binding sites of COO- groups in hydrolysed protein. It reduces the pH of milk rapidly and equilibrates the medium for the growth of Lactobacillus delbrueckii subsp. bulgaricus during yoghurt fermentation. Efforts to increase the diversity of artisanal yoghurt starters are not only important to bring new strains with novel and desirable characteristics, but also for the preservation of natural diversity which diminishes with the overuse and spread of industrial starters to natural resources. In the present study, 14 artisanal yoghurt samples were processed for the isolation of promising strains for yoghurt starter culture production and 66 strains were subsequently characterized. They were all identified as S. thermophilus using species-specific PCR and 16S rRNA gene sequencing. Genotypic diversity at the strain level was investigated by pulsed field gel electrophoresis (PFGE), and 22 homology groups were obtained. Further phenotypic characterization unearthed a significant phenotypic heterogeneity within homology groups, mostly with atypical novel character. Only 7 out of 66 strains showed S. thermophilus type-strain like phenotypic traits. Majority of the isolates were determined to be protease positive and fast milk acidifier to be used as yoghurt starter culture.Article Citation - WoS: 13Citation - Scopus: 14Nanotubular Structures Developed From Whey-Based ?-Lactalbumin Fractions for Food Applications(John Wiley and Sons Inc., 2014) Tarhan, Özgür; Harsa, ŞebnemWhey proteins have high nutritional value providing use in dietary purposes and improvement of technological properties in processed foods. Functionality of the whey-based α-lactalbumin (α-La) may be increased when assembled in the form of nanotubes, promising novel potential applications subject to investigation. The purpose of this study was to extract highly pure α-La from whey protein isolate (WPI) and whey powder (WP) and to construct protein nanotubes from them for industrial applications. For protein fractionation, WPI was directly fed to chromatography, however, WP was first subjected to membrane filtration and the retentate fraction, whey protein concentrate (WPC), was obtained and then used for chromatographic separation. α-La and, additionally β-Lg, were purified at the same batches with the purities in the range of 95%-99%. After enzymatic hydrolysis, WPI-based α-La produced chain-like and long nanotubules with ∼20 nm width while WPC-based α-La produced thinner, miscellaneous, and fibril-like nanostructures by self-assembly. Raman and FT-IR spectroscopies revealed that α-La fractions, obtained from both sources and the nanostructures, developed using both fractions have some structural differences due to conformation of secondary structure elements. Nanotube formation induced gelation and nanotubular gel network entrapped a colorant uniformly with a transparent appearance. Dairy-based α-La protein nanotubules could be served as alternative gelling agents and the carriers of natural colorants in various food processes.Article Citation - WoS: 3Citation - Scopus: 5Production of Food Grade Ss-Galactosidase From Artisanal Yogurt Strains(Taylor and Francis Ltd., 2010) Tarı, Canan; Üstok, Fatma Işık; Harsa, ŞebnemSuperior artisanal isolates of thermophilic lactic acid bacteria producing high lactic acid and β-galactosidase were isolated from traditional Turkish yogurt samples from the Toros mountain region from a highly bio-diverse environment. A full factorial statistical design, with the factors of types of strains and medium formulations under static and agitation conditions, were applied to investigate the effects on β-galactosidase and lactic acid production. Streptococcus thermophilus 95/2 and Lactobacillus delbrueckii subsp. thermophilus 77 exhibited remarkable potential as promising starter culture candidates valuable to various applications in the dairy industry. The efficiency of cell disruption methods was investigated on the extraction of intracellular β-galactosidase enzyme. Lysozyme enzyme treatment was determined as the most effective method, which resulted in approximately 1.5 and 10 times higher enzyme activity than glass bead and homogenization treatment, respectively. © Taylor & Francis Group, LLC.Article Citation - WoS: 48Citation - Scopus: 59Biochemical and Thermal Properties of Ss-Galactosidase Enzymes Produced by Artisanal Yoghurt Cultures(Elsevier Ltd., 2010) Üstok, Fatma Işık; Tarı, Canan; Harsa, Şebnemβ-Galactosidases, produced by pure and mixed cultures of Streptococcus thermophilus 95/2 (St 95/2) and Lactobacillus delbrueckii ssp bulgaricus 77 (Lb 77) isolated from the Toros mountain region of Turkey, were characterised with respect to their biochemical and thermal properties. Optimum pH and temperature for maximum activity were determined and these enzymes were stable in the pH range 7-9 and in the temperature range 20-37 °C, retaining 80-90% of their initial activities. The inactivation energies of β-galactosidase from Lb 77, St 95/2 and mixed culture (Lb 77 and St 95/2) were 51.3, 44.0 and 48.3 kcal mol-1, respectively. Moreover, thermodynamic (ΔG, ΔS, ΔH) and kinetic constants (Km and Vmax) were determined and effects of metal ions were investigated. As a result, these enzymes could be considered as potential candidates for lactose hydrolysis of milk and milk products. © 2009 Elsevier Ltd. Antimicrobial susceptibility testing against 21 antibiotics was performed by agar disk diffusion method, and those isolates resistant to any antibiotic were further analyzed to determine minimum inhibitory concentration by E-test and polymerase chain reaction analysis of vanA and mecA genes. According to disk diffusion test results, a total of 139 strains were resistant to at least one tested antibiotic, with 39 (25.3%) strains being multidrug resistant (MDR) and the other 15 strains being susceptible to all antibiotics. Penicillin G, linezolid, erythromycin, and tetracycline took up 71.4%, 23.4%, 18.2%, and 15.6% of the tested strains, respectively. In addition, all of the strains were susceptible to vancomycin, oxacillin, cefoxitin, and imipenem. Only one strain (S158B) was resistant to both teicoplanin and cefazolin. On the other hand, the presence of vanA and mecA genes was not detected in the strains. Pulsed-field gel electrophoresis analysis was used to identify genetic-relatedness of the MDR strains. It is noteworthy that some strains from different sources showed 100% homology; however, some of MDR strains were found unrelated with 60% or less homology. The high diversity observed in pulsed-field gel electrophoresis results indicated the possible contamination of S. aureus from different sources and routes.
