    Prevalence, Virulence Characterization, and Genetic Relatedness of Listeria Monocytogenes Isolated From Chicken Retail Points and Poultry Slaughterhouses in Turkey

    Listeria monocytogenes is one of the most important foodborne pathogens and is a causal agent of listeriosis in humans and animals. The aim of this study was to determine the prevalence, serogroups, antibiotic susceptibility, virulence factor genes, and genetic relatedness of L. monocytogenes strains isolated from 500 poultry samples in Turkey. The isolation sources of 103 L. monocytogenes strains were retail markets (n = 100) and slaughterhouses (n = 3). L. monocytogenes strains were identified as serogroups 1/2a-3a (75.7%, lineage I), 1/2c-3c (14.56%, lineage I), 1/2b-3b-7 (5.82%, lineage II), 4a-4c (2.91%, lineage III), and 4b-4d-4e (0.97%, lineage III). Most of the L. monocytogenes strains (93.2%) were susceptible to the antibiotics tested. PCR analysis indicated that the majority of the strains (95% to 100%) contained most of the virulence genes (hylA, plcA, plcB, prfA, mpl, actA, dltA, fri, flaA inlA, inlC, and inlJ). Pulsed-field gel electrophoresis (PFGE) demonstrated that there were 18 pulsotypes grouped at a similarity of >90% among the strains. These results indicate that it is necessary to prevent the presence of L. monocytogenes in the poultry-processing environments to help prevent outbreaks of listeriosis and protect public health.

    Is Routine Histopathological Gallbladder Examination Necessary After Cholecystectomy? Evaluation of the Results of 1366 Cholecystectomy Specimens in Single Center

    Objective: It was aimed to evaluate the results of routine histopathological examination after cholecystectomy and to investigate the necessity of routine histopathologic examination after cholecystectomy. Methods: The study was designed retrospectively. One thousand three hundred sixty six patients who underwent laparoscopic and open cholecystectomy at Private Sani Konukoglu Hospital with pre-diagnosis of benign gallbladder disease between November 2011 and May 2017 were included in the study. Patients' demographic data, pathologic results, macroscopic appearance of the specimen, and cancer staging were recorded. The distribution and frequency of pathologic diagnoses and the prevalence of incidental gallbladder cancer (GBC) were evaluated. Pathologic findings were compared in terms of age groups and gender relations. Results: The number of patients included in the study was 1366. Diagnosed with chronic cholecystitis patients were 1,303 (95%), 39 (3%) with acute cholecystitis, 7 (0.5%) with gallbladder cancer, and 17 (1.5%) with other diagnoses of the patients. Statistical significance was found between the groups in terms of the mean age (p = 0.0002). Comparisons between groups in terms of cholesterolysis were statistically significant (p = 0.0003). There was a significant relationship between mucosa atrophy and gender (p = 0.001). Conclusions: The histopathological spectrum of gallbladder is quite extensive. Incidental GBC may not be detected by preoperative imaging methods. Incidental GBC are usually asymptomatic. T2, T3 and T4 GBC were also encountered in our study. All of these patients need additional operations. In the absence of routine histopathologic examination, metastatic advanced GBC may be encountered because no treatment plans could make. Thus, we do recommend routine histopathological examination.

    Evaluation of Hemocytometer Parameters as Potential Biomarkers in Benign Multinodular Goiter and Papillary Thyroid Carcinoma

    Background: Hemocytometer parameters can be important biomarkers for some types of cancers and diseases. There is a need to evaluate their biomarker potential in thyroid diseases. Objectives: The current study aimed at contributing to potential biomarker researches to diagnose papillary thyroid carcinoma (PTC) and benign multinodular goiter (BMG), evaluate the role of these markers to determine the PTC characteristics and contribute to clarification of controversial issues. Methods: The study was designed as a retrospective observational study. The study included 389 patients who underwent total thyroidectomy at private Sani Konukoglu hospital (Sanko University, School of Medicine, Gaziantep, Turkey) due to BMG or PTC diagnosis from November 2011 to May 2017. The study subjects were divided into 3 groups. Groups 1, 2, and 3 were subjects with BMG, PTC, and the control, respectively. The comparisons were made among the groups in terms of hemocytometer parameters. In the group with the thyroid papillary carcinoma diagnosis, the relationship between the hemocytometer parameters and the bilateral tumor presence, single-sided multicentrite, and tumor diameter (size) were investigated. Results: Red cell distribution width (RDW) levels were 15.50 +/- 2.39, 15.68 +/- 2.16, and 12.5 +/- 1.51 in the BMG, PTC, and control groups, respectively. Mean platelet volumes (MPV) were 7.97 +/- 1.19, 8.05 +/- 1.20, and 7.23 +/- 1.39 in the BMG, PTC, and control groups, respectively. MPV and RDW values were significantly lower in group 3 compared with the groups 1 and 2 (P < 0.0001). Plateletcrit (PCT) values were 0.22 +/- 0.05, 0.23 +/- 0.06, and 0.19 +/- 0.05 in the BMG, PTC, and control groups, respectively. PCT was significantly lower in the group 3 compared with the groups 1 and 2 (P = 0.0001). In PTC group, no significant relationship was observed between any of the examined hematological parameters in terms of multicentric and bilateral tumor presence. Conclusions: RDW, MPV, haemoglobin content, and PCT were significantly higher in the BMG and PTC groups. This increase was not specific for either of the 2 groups. No significant difference was found between any of the hemocytometer parameters and the multicentricity and bilaterality of PTC. But, a possible correlation was observed between activated partial thromboplastin time (aPTT), hemoglobin content, and tumor size.

    Assessing the Impact of Non-Thermal and Thermal Treatment on the Shelf-Life of Onion Juice

    Onion (Allium cepa L.) juice is a marinating agent for meat and fish marination and readily usable sauce for any meal that has onion in its formulation. This study aims to assess the microbiological and physicochemical changes in the onion juice processed by UV-C irradiation (0.5 mm sample depth, 30 min exposure time, 7.5 mW/cm(2) UV incident intensity) and conventional heat treatment (74.5 degrees C, 12 min) during its storage. Microbiological results showed processing by UV-C irradiation or heat treatment under optimum conditions extended the microbial shelf-life of untreated onion juice by minimum 6-times. Total colour change of heat-treated samples was lower than that of untreated and UV-C treated samples for 12 weeks. Also, pH, total titratable acidity, total soluble solids content, turbidity, NEBI and total phenolic content were monitored for 12 weeks. The results of this study will form scientific infrastructure for onion juice manufacturers to decide on the processing method with respect to its shelf-life.

    Mid-Infrared Spectroscopic Detection of Sunflower Oil Adulteration With Safflower Oil

    The oil industry is in need of rapid analysis techniques to differentiate mixtures of safflower-sunflower oils from pure oils. The current adulteration detection methods are generally cumbersome and detection limits are questionable. The aim of this study was to test the capability of a mid-infrared spectroscopic method to detect the adulteration of sunflower oil with safflower oil compared to fatty acid analysis. Mid-infrared spectra of pure oils and their mixtures at the 10-60% range were obtained at 4000-650 cm(-1) wavenumber and fatty acid profiles were determined. Data were analyzed by multivariate statistical analysis techniques. The lowest level of detection was obtained with mid-infrared spectroscopy at 30% while the fatty acid profile could determine adulteration at around 60%. Adulteration levels were predicted successfully using PLS regression analysis of infrared data with R-2 (calibration) = 0.96 and R-2 (validation) = 0.93. As a rapid and minimum waste generating technique, mid-infrared spectroscopy could be a useful tool for the screening of raw material to detect safflower-sunflower oil mixtures.

    Chemometric Analysis of Chemo-Optical Data for the Assessment of Olive Oil Blended With Hazelnut Oil

    The main objective of this study was to determine different hazelnut oil concentrations in extra virgin olive oil (EV00) belonging to different geographical regions inside Turkey using the combination of a SAW sensor based electronic nose (e-nose) and a machine vision system (MVS). We leveraged the oil characterisation given by the two easy-to-use and complementary experimental techniques through the adoption of conventional PCA for data exploration and random forests (RF) for supervised learning. The e-nose/MVS combination allows significantly better results both in adulteration detection independently of EVOO's geographical provenance and in EVO0 geographical provenance determination, independently of the adulteration level, with respect to the single characterisation method. RF analysis also produces feature ranking, permitting to shed light on which oils' characteristics influence the learning result. We found that EV00 geographical provenance discrimination is mainly due to yellowness and guaiacol content, while (E)-2-hexenal chiefly determines the prediction of the hazelnut level.

    Detection of Listeria Species in Fresh Fish and Fish Market Environment by Ims Technique in Turkey

    The incidence of Listeria spp. was investigated in fresh fish (n = 100) sold at retail markets and in the environmental and personnel samples (n = 100) obtained from several fish markets in Afyonkarahisar, Turkey by immunomagnetic separation technique. The fish samples analysed included anchovy, trout, carp and grey mullet (25 of each). Six (6 %) of the fish samples were found positive for Listeria spp. and the overall incidence of Listeria spp. was 10 % in the environmental and personnel samples. Three Listeria spp., Listeria monocytogenes, L. ivanovii, L. grayi were recovered from the samples examined. In addition, L. seeligeri from a fish sample and L. innocua from an environmental sample (box) were isolated. It was found that L. monocytogenes was only detected in fresh water fish with an incidence of 8 %. For the environmental samples, knives and refrigerators had the highest (20 %) and personnel samples had the least (5%) levels of contamination. It can be concluded that the fish sold at retail markets are moderately contaminated with various species of Listeria including L. monocytogenes, which may pose a risk for human health. The presence of these bacteria in environmental and personnel samples of fish markets is also an important possible sources of cross-contamination of the fish and other seafood. The results of this study indicate the necessity of the implementation of good hygiene and sanitary practices in order to prevent and/or reduce the contamination of fishery products by Listeria spp. at the retail level.

    Capsaicin Emulsions: Formulation and Characterization

    Oleoresin capsicum, the oil extract of chili pepper, is mainly composed of capsaicin. Capsaicin is a hydrophobic volatile compound exhibiting antimicrobial activity against various microorganisms. Capsaicin in the form of an emulsion-based carrier system could be a good alternative to enhance bioavailability and simultaneously to increase the shelf-life of food. In this study, capsaicin emulsions were formulated using three different surfactants (Tween 80, commercial soy lecithin, and sucrose monopalmitate/SMP). Effects of aqueous phase composition, pH, and heating the pre-homogenized dispersion were investigated. For characterization, NMR relaxometry, color, turbidity, and antioxidant activity experiments were conducted. Antimicrobial efficacies of the emulsions were also evaluated against Escherichia coli andStaphylococcus aureus. Mean particle sizes of emulsions with surfactants Tween 80, lecithin, and SMP were found to be 68.30, 582.63, and 50.10 nm, respectively. Lecithin-containing emulsions showed the highest antimicrobial activity against S. aureus with 4.60 log reduction, whereas the same effect was observed in Tween 80-containing emulsions against E. coli with 3.86 log reduction. Emulsions prepared with SMP showed the highest antioxidant activity with 0.482 mg DPPH/L emulsion. The formulated emulsions have the potential to be used in food industry as antimicrobial food grade solutions.

    Usage of Rice Milk in Probiotic Yoghurt Production

    There are previous studies focusing on the production of probiotic and fermented dairy products made using vegetable based raw materials like oats and soy, however there is a limited number of studies on the usage of rice milk in fermented dairy products. Four different types of yoghurt samples were produced and stored for 21 days at 4°C. Physical, chemical, microbiological and sensory characteristics of the samples were performed at the 1st, 7th, 14th and 21st days of the storage. It was determined that rice milk increased the viscosity values but decreased the values of the texture, whey separation and the chemical and microbiological properties of yoghurts. Acetaldehyde, acetoin, acetone and diacetyl of carbonyl compounds were detected as main flavor components of yoghurt samples. In the sensory analysis, scores decreased as the rice milk proportions in yoghurt was increased and the panelists reported that P1 sample (25% rice milk + 75% cow's milk) was the closest sample to the control sample (100% cow's milk). Generally speaking, samples containing rice milk did not give good results. However, P1 samples were the most favored products among the samples containing rice milk as they were the closest product to the control group. The consumption of such products is continuously increasing as the customers' tendency to consider them as functional products rather than traditional food products increase.

    Kinetic and Structural Characterization of Interaction Between Trypsin and Equisetum Arvense Extract

    Objective: In this study the inhibitory effect of E. arvense extract on trypsin activity and the effect of trypsin on E. arvense extract were studied. In addition the nature of the interaction between the extract and trypsin was investigated. Methods: The inhibitory effect ethanol extract of E. arvense on trypsin activity was determined using trypsin enzyme assay. The structural effects of the extract-trypsin interaction for the extract were analyzed by FTIR. Finally, the HPLC analyses were carried out to analyze the individual components of the extract and the supernatant and soluble precipitate phases. Results: E. arvense extract was found to decrease total percent activity of trypsin to 5% in 24 hour at 24 °C. FTIR analyses indicated that the interaction between trypsin and E. arvense extract caused changes in the structure and hydrogen bonding behavior and composition of the extract proteins. These interactions also caused the extract lipids to accumulate in the insoluble precipitate phase. Most of the phenolics remained in the supernatant phase enhancing the inactivation of trypsin. However, the precipitated compounds were shown to be of apolar in nature as shown in the HPLC chromatograms. Conclusion: The methods that were used showed that the high phenolic content of E. arvense was the main reason for the inhibition of trypsin enzyme activity by denaturing the enzyme.
