    Mitotyping of Random Bred Cats and Pure Breed Cats (turkish Angora and Turkish Van) Using Non-Repetitive Mitochondrial Dna Control Region

    The Fertile Crescent appears to be the most plausible region where the domestication of cats commenced through a mutually beneficial relationship between wild cats and early agrarian societies. These domesticated cats then journeyed across the globe mirroring the paths of human migration. An examination of mitochondrial DNA (mtDNA) control region-based mitotyping suggested that a significant majority, exceeding 80%, of globally sampled random-bred and pure-bred cats could be categorized into 12 predominant mitotypes. However, the extent of mitotype diversity within random-bred cats from regions proximate to the Fertile Crescent remains inadequately explored. In light of this we aimed to investigate the mitotype diversity in random bred cats sampled from various regions across Turkey. Additionally, we sought to establish a comparison with the mito-type profiles of locally recognized pure breeds, namely the Turkish Angora and Turkish Van. To unravel their evolutionary narratives, we engaged in comprehensive population genetics analyses at both the individual and mitotype-based levels. Our study encompassed a sample size of 240 specimens, forming the basis for both mitotyping and population genetics scrutiny. Our analysis yielded the identification of nine 'universal' mitotypes (A-J), alongside an 'outlier' mitotype group I. Notably mitotypes A and D emerged as particularly prevalent in contrast to the lesser occurrence mitotypes C, G, and H. With the realm of random bred cats the structure of haplotypes exhibited remarkable diversity presenting distinctions from Turkish Angora and Van breeds. Nucleotide diversity was higher compared to previous reports from Turkey and was one of the highest among reported world cat population estimates. Intriguingly, our investigations did not unveil any pronounced instances of strong selection, population expansions or contractions within any specific population or mitotype. To conclude, our study represents a pioneering effort in uncovering the mitotype profiles and haplotype structures inherent to both random-bred and pure breed cats in Turkey. This endeavor not only broadens our understanding of the feline genetic landscape within the region but also lays the foundation for future inquiries into the evolutionary trajectories and genetic legacies of these feline populations.

    A Screen-Printed Electrode Modified With Gold Nanoparticles/ Cellulose Nanocrystals for Electrochemical Detection of 4,4'-methylene Diphenyl Diamine

    Developing simple, cost-effective, easy-to-use, and reliable analytical devices if of utmost importance for the food industry for rapid in-line checks of their products that must comply with the provisions set by the current legislation. The purpose of this study was to develop a new electrochemical sensor for the food packaging sector. More specifically, we propose a screen -printed electrode (SPE) modified with cellulose nanocrystals (CNCs) and gold nanoparticles (AuNPs) for the quantification of 4,4'-methylene diphenyl diamine (MDA), which is one of the most important PAAs that can transfer from food packaging materials into food stuffs. The electrochemical performance of the proposed sensor (AuNPs/CNCs/SPE) in the presence of 4,4'- MDA was evaluated using cyclic voltammetry (CV). The modified AuNPs/CNCs/SPE showed the highest sensitivity for 4,4'-MDA detection, with a peak current of 9.81 mu A compared with 7.08 mu A for the bare SPE. The highest sensitivity for 4,4'-MDA oxidation was observed at pH = 7, whereas the detection limit was found at 57 nM and the current response of 4,4'-MDA rose linearly as its concentration increased from 0.12 mu M to 100 mu M. Experiments using real packaging materials revealed that employing nanoparticles dramatically improved both the sensitivity and the selectivity of the sensor, which can be thus considered as a new analytical tool for quick, simple, and accurate measurement of 4,4 '-MDA during converting operations.

    Detection of Vinegar Adulteration With Spirit Vinegar and Acetic Acid Using Uv–visible and Fourier Transform Infrared Spectroscopy

    Vinegar is one of the commonly adulterated food products, and variations in product and adulterant spectrum make the detection of adulteration a challenging task. This study aims to determine adulteration of grape vinegars with spirit vinegar and synthetic acetic acid using different spectroscopic methods. For this purpose, grape vinegars were mixed separately with spirit vinegar and diluted synthetic acetic acid (4%) at 1–50% (v/v) ratios. Spectra of vinegars and mixtures were obtained with UV–visible and Fourier-transform infrared (FTIR) spectrometers. Data were evaluated with various chemometric methods and artificial neural networks (ANN). Correct classification rates of at least 94.3% and higher values were obtained by the evaluation of both spectroscopic data along with their combination with chemometric methods and ANN for discrimination of non-adulterated and adulterated vinegars. UV–vis and FTIR spectroscopy can be rapid and accurate ways of detecting adulteration in vinegars regardless of adulterant type.

    Shelf Life Extension of Strawberry Juice by Equivalent Ultrasound, High Pressure, and Pulsed Electric Fields Processes

    Nonthermal processing technologies have focused on the production of safe, fresh-like and high quality food products very much in line with current consumer demands. It is a high priority to maintain the quality attributes of the food during its shelf life. In this study; microbial stability, physicochemical properties and phytochemical characteristics of strawberry juice (SJ) pasteurized by ultrasonication (US) (55 degrees C, 0.29 W/mL acoustic energy density, 120 mu m amplitude, 3 min), high pressure processing (HPP) (300 MPa, 1 min), and pulsed electric fields (PEF) (35 kV/cm, 27 mu s) were evaluated during 42 days of storage at 4.C in comparison with conventional thermal pasteurization as a reference treatment (72 degrees C, 15 s). The nonthermal processes were equivalent in terms of E. coli inactivation since the selected processing conditions previously led to almost identical inactivation level (at least 5-log) of inoculated E. coli. Thus, the current study demonstrates how these equivalent US, HPP, and PEF treatments differ from each other in terms of their effect on SJ natural microbiota and quality characteristics during refrigerated storage. Results showed that US, HPP, and heat treatment ensured the microbial stability of SJ for at least 42 days while PEF extended the shelf life of SJ by at least 28 days based on the natural microbiota. No significant difference was found for the total soluble solids of the processed samples (p > 0.05) whereas acidity and pH of the samples varied during the storage period (p < 0.05). Immediately after processing, the total phenolic contents and antioxidant activities of SJ were better retained by HPP and PEF compared to thermal pasteurization. Furthermore, HPP and PEF significantly increased total anthocyanin content of SJ by 15 and 17% with respect to untreated SJ (p < 0.05). Phytochemical characteristics of processed SJ started to decrease after 7 days of storage irrespective of treatment type. HPP treated juices showed significantly higher levels of total anthocyanin and antioxidant activity at the final day of storage. Principal component and cluster analysis showed that the processed SJ samples had higher similarity to the untreated fresh SJ during storage up to 14 days, while the samples beyond this storage period clustered together and discriminated from the rest indicating a decreased similarity to the fresh juice. This study rendered simultaneous evaluation of several quality characteristics during storage of pasteurized strawberry juice based on the equivalent processing approach and multivariate data analysis. Under the selected processing conditions, HPP was the best option to extend the shelf life of SJ and enhance its phytochemical characteristics.

    Primary Surgery With Systemic Therapy in Patients With De Novo Stage Iv Breast Cancer: 10-Year Follow-Up; Protocol Mf07-01 Randomized Clinical Trial

    Background: The aim of this randomized clinical trial was to evaluate the overall survival (OS) data of patients diagnosed with de novo stage IV breast cancer (BC) who received locoregional treatment (LRT) over a 10-year follow-up. Study Design: The MF07-01 is a 1:1 multicenter, randomized clinical trial comparing the LRT with systemic therapy (ST), where ST was given to all patients either immediately after randomization or after surgical resection of the intact primary tumor. Results: A total of 278 patients were randomized and 265 patients were in the final analysis. At 10-year follow-up, survivals were 19% (95% CI 13%–28%) and 5% (95% CI 2%–12%) in the LRT group and ST group, respectively. Median survival was 46 months for the LRT group and 35 months for the ST group, and hazard of death was 29% lower in the LRT group compared with the ST group (hazard ratio [HR] 0.71; 95% CI 0.59–0.86; p = 0.0003). Conclusions: Patients with a diagnosis of de novo stage IV BC who underwent LRT followed by ST had a 14% higher chance of OS by the end of the 10-year follow-up compared with the patients who received only ST. The longer study follow-up revealed that LRT should be presented to patients when discussing treatment options. © 2021 American College of Surgeons

    Comparison of Radio-Guided Occult Lesion Localization (roll) and Magnetic Occult Lesion Localization (moll) for Non-Palpable Lesions: a Phantom Model Study

    We sought to compare radio-guided localization and magnetic tracer localization techniques by using a phantom model for excision of nonpalpable breast lesions. There was no difference between the groups in the duration of operative excision, specimen weight, or specimen volume. Magnetic occult lesion localization can be performed in clinics without the need for a nuclear medicine team and radiation safety procedures. Background: Localization of nonpalpable breast cancers can be achieved with several techniques. We sought to compare radio-guided localization (ROLL) and magnetic tracer localization (MOLL) techniques by using a phantom model we previously developed, which can provide an accurate simulation for excision of nonpalpable breast lesions. Materials and Methods: We designed 20 phantom models (10 MOLL, 10 ROLL group) for localization. A handheld gamma probe for the ROLL group and a manual magnetometer (SentiMag) for the MOLL group were used to test the ability of the modality to detect olives in turkey breasts. The excision time for each procedure, specimen size, and weight of the specimens removed from the turkey breasts were recorded. Results: Both techniques resulted in 100% retrieval of the lesions. There was no difference between the groups in the duration of operative excision, specimen weight, or specimen volume. Conclusion: This experimental trial found similar success rates for ROLL and MOLL in localization of occult lesions using the turkey breast phantom model. MOLL can be performed in clinics without the need for a nuclear medicine team and radiation safety procedures. (C) 2019 Elsevier Inc. All rights reserved.

    Use of Ftir and Uv-Visible Spectroscopy in Determination of Chemical Characteristics of Olive Oils

    It was aimed to predict fatty acid ethyl ester (FAEE), wax, diacylglycerol (DAG) and color pigment contents of olive oils by using rapid and non-destructive spectroscopic techniques (FTIR and UV-vis) individually and in combination. Prediction models were constructed by using partial least squares (PLS) regression with cross and external validation. FAEEs were estimated best with FTIR + UV-Vis spectroscopy (R-cv.(2) = 0.84, R-pred(2) = 0.90, and RPD = 3.0). PLS model with R-cv.(2) = 0.79, R-pred(2) = 0.71, and RPD = 1.9 was obtained for the estimation of 1,2 DAG using FTIR spectral data. Major pigments, lutein, pheophytin a and their derivatives and total xanthophylls were quantified successfully by FTIR + UV-Vis with a range of R-cv.(2) of 0.71-0.85, R-pred(2) of 0.70-0.84, and RPD = 1.5-2.5 values but the prediction of the rest of the pigments were poor (R-cv(2) = 0.60-0.76, R-pred(2) = 0.42-0.62, and RPD = 1.2-1.5). Combination of two spectral data resulted in average prediction of wax content of oils (R-cal(2) = 0.95, R-pred(2) = 0.75, and RPD = 1.9). FTIR and UV-vis spectroscopic techniques in combination with PLS regression provided promising results for the prediction of several chemical parameters of olive oils; therefore, they could be alternatives to traditional analysis methods.

    Hot Nasal Packing With Hot Saline Irrigation for Hemostasis After Adenoidectomy: a Prospective Randomized Controlled Study

    Objective: This study aimed to investigate the efficacy of hot posterior nasal packing and hot saline irrigation in bleeding control after adenoidectomy. Methods: A total of 130 patients scheduled for adenoidectomy were included in the study, and randomized into two groups at the beginning of the surgical operation. After adenoidectomy, saline impregnated tampon, and saline irrigation at room temperature (22 degrees C) was applied to the patients in one group while 50 degrees C saline impregnated tampon and saline irrigation at the same temperature were applied to the patients in the other group. We recorded hemostasis for up to 3 min after tamponade for bleeding control, and the amount of bleeding into the nasopharynx. Results: The age of the patients ranged from 1.5 to 13 years (mean +/- SD: 6.07 +/- 3.08 years, and 5.33 +/- 2.55 years, 22 degrees C and 50 degrees C saline irrigation groups, respectively). There were 37 males and 28 females in the 22 degrees C saline group, while 34 males and 31 females in the 50 degrees C saline group. When comparing the two groups, there was no statistically significant difference in terms of duration of hemostasis (p = 0.64). However, bleeding scores at 2 nd min after the tamponade were significantly lower in the 50 degrees C saline group (p = 0.007). The amount of bleeding in the 50 degrees C saline group was also significantly lower than the 22 degrees C saline group (p = 0.015). Conclusion: In this study, application of 50 degrees C saline impregnated tampon, and hot saline irrigation was found to be more effective in the control of bleeding after adenoidectomy by reducing the amount of bleeding compared to 22 degrees C saline impregnated tampon application and saline irrigation at 22 degrees C. However, hot nasal packing and hot saline irrigation did not affect duration of hemostasis and cauterization.

    Decontamination of Seeds Destined for Edible Sprout Production From Listeria by Using Chitosan Coating With Synergetic Lysozyme-Nisin Mixture

    This study aimed at decontamination of seeds destined for edible sprout production from Listeria using chitosan (CS) coatings incorporated with synergetic lysozyme-nisin (LYS-NIS) mixtures. Low molecular weight (LMW) CS coating showed the highest potency against Listeria innocua, followed by medium molecular weight (MMW) and high molecular weight (HMW) CSs. The LMW CS film with LYS-NIS also caused almost 1.5-fold greater log reduction (similar to 5 log) in initial L. innocua load of broth culture than MMW and HMW CS films with LYS-NIS within 6 days. Moreover, LMW CS coating with LYS-NIS reduced the initial Listeria loads of inoculated mung beans, lentils, and wheats by 3.3, 3.4 and 4.1 log, respectively. Antimicrobial coating did not affect seed germination rates considerably. The LYS-NIS addition increased yellowness and opacity of films, and caused limited changes in their mechanical and morphological properties. LMW CS coating with LYS-NIS reduces risk of listeriosis from sprouted seeds.

    Utilization of Xylan-Type Polysaccharides in Co-Culture Fermentations of Bifidobacterium and Bacteroides Species

    Although most members of the genus Bifidobacterium are unable to utilize xylan as a carbon source, the growth of these species can be induced by this polysaccharide in the gut environment. This indicates a requirement for an association between Bifidobacterium species and some other members of gut microbiota. In this study, the role of cross-feeding between Bifidobacterium and Bacteroides species in the bifidogenic effect of xylan was investigated using in-vitro pure and co-culture fermentations. The pure culture studies showed that among the Bifidobacterium species tested, only Bifidobacterium animalis subsp. lactis was able to utilize xylooligosaccharides. The co-culture of this strain with Bacteroides species enabled it to grow in the presence of xylan. These results suggest that the ability of Bacteroides species to hydrolyze xylan could allow the proliferation of specific Bifidobacterium species in the gut through substrate cross-feeding.
