Article Effect of Marination on the Formation of Polycyclic Aromatic Hydrocarbons in Grilled Vegetables(Wiley, 2025) Kacmaz Ozcetin, Sibel; Artok, LeventThe effect of marination on the formation of polycyclic aromatic hydrocarbons (PAH) in charcoal-grilled vegetables was studied. Various marinade ingredients, including apple cider vinegar, red grape vinegar, lemon juice, garlic powder, black pepper, and the food additive tert-butylhydroquinone (TBHQ) were applied to vegetable samples before charcoal grilling. The total phenolic content (TPC) and total antioxidant capacity (TAC) of each marinade ingredient were assessed for their contribution to PAH inhibition. A substantial decrease in PAH4 formation was observed in marinated vegetables. Red grape vinegar exhibited the strongest average inhibitory effect on total PAH4 formation (75%), followed by apple vinegar (68%), lemon juice (52%), garlic powder (34%), and black pepper (30%). Additionally, the TBHQ (67%) demonstrated a strong inhibitory effect, reducing total PAH4 formation by 67%. First, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide activity assay studies of two isoindole-1,3-dione derivatives were performed against A549 cell lines. Both compounds showed inhibitory effects on the viability of A549 cells. Then, we explored the potential of these compounds as active ingredients by in vivo studies. Nude mice were given A549-luc lung cancer cells, and tumor growth was induced with a xenograft model. Then, nude mice were divided into three groups: the control group, compound 3 group, and compound 4 group. After application of each compound to the mice, tumor sizes, their survival, and weight were determined for 60 days. Furthermore, toxicological studies were performed to examine the effects of the drugs in mice. In addition to toxicological studies, histopathological analyses of organs taken from mice were performed, and the results were evaluated. The obtained results showed that both N-benzylisoindole derivatives are potential anticancer agents.Article Citation - WoS: 9Citation - Scopus: 12Effects of Pomegranate and Pomegranate-Apple Blend Juices on the Growth Characteristics of Alicyclobacillus Acidoterrestris Dsm 3922 Type Strain Vegetative Cells and Spores(Elsevier Ltd., 2015) Molva, Çelenk; Baysal, Ayşe HandanThe present study examined the growth characteristics of Alicyclobacillus acidoterrestris DSM 3922 vegetative cells and spores after inoculation into apple, pomegranate and pomegranate-apple blend juices (10, 20, 40 and 80%, v/v). Also, the effect of sporulation medium was tested using mineral [Bacillus acidoterrestris agar (BATA) and Bacillus acidocaldarius agar (BAA)] and non-mineral containing media [potato dextrose agar (PDA) and malt extract agar (MEA)]. The juice samples were inoculated separately with approximately 105CFU/mL cells or spores from different sporulation media and then incubated at 37°C for 336h. The number of cells decreased significantly with increasing pomegranate juice concentration in the blend juices and storage time (p<0.001). Based on the results, 3.17, 3.53, and 3.72 log cell reductions were observed in 40%, 80% blend and pomegranate juices, respectively while the cell counts attained approximately 7.17logCFU/mL in apple juice after 336h. On the other hand, the cell growth was inhibited for a certain time, and then the numbers started to increase after 72 and 144h in 10% and 20% blend juices, respectively. After 336h, total population among spores produced on PDA, BATA, BAA and MEA indicated 1.49, 1.65, 1.67, and 1.28 log reductions in pomegranate juice; and 1.51, 1.38, 1.40 and 1.16 log reductions in 80% blend juice, respectively. The inhibitory effects of 10%, 20% and 40% blend juices varied depending on the sporulation media used. The results obtained in this study suggested that pomegranate and pomegranate-apple blend juices could inhibit the growth of A. acidoterrestris DSM 3922 vegetative cells and spores.Article Citation - WoS: 19Citation - Scopus: 23Inhibition of Catechol-O (comt) by Some Plant-Derived Alkaloids and Phenolics(Elsevier Ltd., 2010) Yalçın, Dilek; Bayraktar, OğuzIn this study, as an alternative to the medicines, natural compounds extracted from plant species (Peganum harmala, Cistus parviflorus and Vitex agnus-cactus) were investigated in order to inhibit the catechol-O-metyhltranferase (COMT) activity. In fluorometric enzyme assay, S-adenosylmethionine (SAM) and aesculetin (ES) were used as methyl donor and acceptor substrates, respectively. Their Km values were determined as 3.5 ± 0.3 μM and 6.4 ± 0.4 μM in absence of inhibitor. Inhibition performances of the plant-derived polyphenolics and alkaloids were determined. Inhibitory effect of alkaloids extracted from P. harmala seeds was found the highest among the plant extracts; however, it was lower than that of 3,5-DNC. In case of inhibition mechanism, mixed type inhibition was observed for alkaloid extract whereas uncompetitive inhibition was observed for 3,5-DNC. In case of polyphenolic extracts obtained from C. parviflorus and V. agnus-cactus leaves, mechanism were also explained as mixed type inhibition and their αKi values were calculated as 1.99 ± 0.35 μg/ml and 9.48 ± 0.58 μg/ml, respectively. © 2009 Elsevier B.V.
